中文摘要: 针对牛奶中恩诺沙星的代谢物环丙沙星残留的问题,该研究开发一种胶体金试纸条快速检测牛奶中的环丙沙星残留。研究制备环丙沙星包被抗原,对环丙沙星抗体进行表征,优化胶体金标记条件,选用最佳标记pH 和最佳蛋白添加量,以硝酸纤维素膜为固相载体,环丙沙星包被抗原为检测带(T 带),羊抗兔二抗为质控带(T 带)。依据免疫竞争法原理,建立胶体金免疫层析试纸条快速检测牛奶中环丙沙星的方法,方法检出限为6.25 ng/mL,对牛奶样品的检出限为12.50 ng/mL。样品前处理方法简单,在10 min 内即可目测判断结果。
Abstract:In order to solve the problem of ciprofloxacin residue which was the major metabolite of enrofloxacin in milk,a colloidal gold test strip for rapid detection of ciprofloxacin residues in milk was established.In this paper,ciprofloxacin coated antigen was prepared and the ciprofloxacin antibody was characterized and the conditions of colloidal gold labeling were optimized. The optimal labeling pH and the optimal protein addition were selected.The nitrocellulose membrane was used as the solid phase carrier with coated antigen as test line and goat anti-rabbit as control line.According to the principle of immuno competition method,a method for rapid detection of ciprofloxacin in milk by colloidal gold immunochromatographic test strip was established.The detection limit of the method was 6.25 ng/mL,and the detection limit of milk samples was 12.50 ng/mL.The sample pretreatment method was simple and the result could be visually judged within 10 minutes.
