原花青素白藜芦醇浆对大鼠酒精性脑损伤的预防性保护作用研究

中文摘要: 以50只SD 大鼠为实验对象,随机均分为正常对照组、模型组、原花青素白藜芦醇浆低、中、高剂量[0.5、1.5、4.5 g(/kg·bw)]干预组。除正常对照组给予等量生理盐水灌胃外,其余各组采用52°白酒灌胃建立酒精性脑损伤大鼠模型,同时灌胃给予相应剂量的原花青素白藜芦醇浆,每天1 次。灌胃8 d 后,通过计算体重增长幅度,检测大鼠血清超氧化物歧化酶(superoxide dismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量,脑组织中SOD、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)和Ca2+-Mg2+-三磷酸腺苷酶(adenosine triphosphate,ATPase)活性,抑制羟自由基能力,MDA、钙离子(Ca2+)含量,分析不同剂量的原花青素白藜芦醇浆对酒精性脑损伤大鼠的作用。结果表明:原花青素白藜芦醇浆能对抗并清除自由基,提高SOD、GSH-Px 和Ca2+-Mg2+-ATP 酶活性,提高抑制羟自由基能力,降低大鼠血清MDA 和脑MDA、Ca2+含量,减轻精神、行为障碍,减弱酒精对食欲和胃肠消化的不利影响,对酒精性脑损伤起到预防性保护作用。
Abstract:Fifty Sprague-Dawley rats were randomly divided into normal group,model group,and low,middle,high dose group of proanthocyanidins resveratrol pulp intervention groups [0.5,1.5,4.5 g/(kg·bw)].Normal group was given normal saline in the same volume while other groups were given 52°liquor into stomach of rats to establish alcoholic brain damage model. Meanwhile,rats in low,middle,high proanthocyanidins resveratrol pulp groups were administered with corresponding foods once a day. After 8 days of gavage,weight gain ranges were calculated,and the activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and Ca2+-Mg2+-ATPase,the suppression ability of hydroxyl free radical,and the contents of malondialdehyde (MDA),Ca2+were detected in brain homogenate,and the activities of SOD and the contents of MDA were also detected in serum. The effects of different doses of proanthocyanidins resveratrol pulp on alcoholic brain damage in rats were analyzed.The results showed that proanthocyanidins resveratrol pulp might have preventive protection effects on alcoholic brain damage by scavenging free radicals,increasing the activity of SOD,GSH-Px,Ca2+-Mg2+-ATPase and the suppression ability of hydroxyl free radical,decreasing the contents of MDA amd Ca2+,alleviating the mental and behavioral disorders,weakening the adverse effects of alcohol on appetite and gastrointestinal digestion.
