中文摘要: 采用响应面法优化黑果枸杞蛋白(Lycium ruthenicum protein,LRP)提取最佳工艺,研究黑果枸杞蛋白的抗衰老活性。在单因素试验基础上,以蛋白提取率为响应值,提取时间、提取次数及液料比为考察因素,采用响应面法对LRP提取工艺进行优化。试验结果表明LRP提取最佳条件为pH 7,提取时间21 h,提取次数2次,液料比17∶1(mL/g);经验证试验得出平均蛋白提取率为8.62%,与模型预测值相近,优化后的提取工艺稳定可行。以D-半乳糖制衰老模型小鼠为研究对象,评价LRP对衰老模型小鼠抗衰老的作用。昆明种小鼠随机分为空白、模型、黑果枸杞蛋白高、中、低剂量组(125、250、500 mg/kg)。给药30 d后,测定衰老模型小鼠血清中谷胱甘肽过氧化物酶(gutathion peroxidase,GSH-Px)、超氧化物歧化酶(superoxide dismutase,SOD)、过氧化氢酶(catalase,CAT)活力和丙二醛(malondialdehyde,MDA)含量。结果显示,LRP能显著降低MDA含量并提高SOD、GSH-Px和CAT活力。结果表明黑果枸杞蛋白对D-半乳糖诱导的衰老小鼠具有延缓衰老作用,其机制可能与清除体内自由基有关。
Abstract:To optimize the extraction process of Lycium ruthenicum protein (LRP)by response surface methodology.The anti-aging activity of LRP was studied.On the basis of single-factor test,extraction time,extraction times and liquid-to-material ratio were selected as investigation factors,the extraction process was optimized through the response surface methodology.The optimal extraction conditions were as follows:pH 7,extraction time 21h,extraction times 2 times,liquid to material ratio 17 ∶1 (mL/g);the average protein extraction rate was 8.62%after validation test,which was close to the model predictive value.The optimized extraction process was stable and feasible.The D-galactose aging model mice were used as the research object to evaluate the anti-aging effect of LRP on aging model mice.Kunming mice were randomly divided into a blank control group,an aging model group,and low-dose,medium-dose,and high-dose group of LRP content(125,250,500 mg/kg,respectively).After 30 days,The activities of superoxide dismutase (SOD)and glutathione peroxidase(GSH-Px),and the contents of malondialdehyde(MDA)and Catalase(CAT)in the serum were assayed.LRP could significantly reduce MDA content and increase SOD,GSH-Px and CAT activity.The results indicated that LRP had a delayed anti-aging effect on D-galactose-induced aging mice,and its mechanism might be related to the elimination of free radicals in vivo.
