中文摘要: 为探索粗甘油综合利用的途径,采用平板培养和摇瓶发酵的方法从4株油脂酵母菌株中筛选出利用粗甘油发酵产油脂的高产菌株,并考察培养基中碳源氮源对菌株发酵产油脂的影响,在此基础上,对菌株的发酵过程进行研究,并对油脂中脂肪酸的组成进行分析。结果表明:斯达油脂酵母ZW-25利用粗甘油发酵产油脂效果较好;培养基中适宜的碳源氮源组成为:粗甘油浓度40 g/L,酵母粉0.5 g/L,(NH4)2SO42.0 g/L,在此条件下,菌株的生物量、油脂产量、油脂含量和油脂系数分别可达13.79 g/L、6.97 g/L、50.52%、18.12;菌株适宜的发酵时间为144 h,油脂中脂肪酸主要是C16和C18系脂肪酸。上述结果显示,斯达油脂酵母ZW-25具有良好的应用潜力,研究结果为粗甘油的综合利用提供了新的途径。
Abstract:Abstract:In order to explore the comprehensive utilization approach of crude glycerol,the high-producing strain of using crude glycerol fermentation for producing lipid was selected in 4 Lipomyces yeast strain by plate culture and shake-flask fermentation,the effect of carbon and nitrogen source in culture medium on fermenting crude glycerol for producing lipid of the selected strain was studied.On the basis,fermenting course of the selected strain was studied,and the fatty acids of lipids were analysed.The results showed that Lipomyces starkeyi ZW-25 was good at fermenting crude glycerol for producing lipid,the suitable carbon and nitrogen source composition in culture medium was such as follows:crude glycerol 40 g/L,yeast powder 0.5 g/L,(NH4)2SO42.0 g/L.The biomass,lipid yield,lipid content and lipid coefficient could reach 13.79 g/L,6.97 g/L,50.52%and 18.12 under these conditions.The suitable time of fermentation for producing lipid by Lipomyces starkeyi ZW-25 was 144 h,and the main fatty acids of lipids were C16and C18fatty acids.These conclusions showed that Lipomyces starkeyi ZW-25 has good application potential,it also provided a new approach for the comprehensive utilization of crude glycerol.
