中文摘要: 以咖啡酸为对照品,采用普鲁士蓝比色法测定蒲公英提取物中总酚酸的含量并对方法学进行考察和优化。并通过单因素考察方法,确定最佳测定方法:显色剂、缓冲剂十二烷基硫酸钠(sodium dodecyl sulfate,SDS)用量为2.0 mL,盐酸浓度为0.1 mol/L,显色20 min。在最佳测定条件下,测得蒲公英提取物中总酚酸含量为120.5 mg/g。该方法稳定性好,精密度高,咖啡酸在2.0 μg/mL~20.0 μg/mL呈现良好的线性关系(r=0.999 2),平均回收率为98.86%,RSD为0.84%;结果表明该方法简单易行,稳定可靠,可作为蒲公英提取物中总酚酸含量的测定方法。
Abstract:The content of total phenolic acids in the extract of Taraxacum mongolicum was determined by Prussian blue colorimetry and the methodology was investigated and optimized,with caffeic acid as the reference material.The dosage of chromogenic agent and sodium dodecyl sulfate(SDS),the concentration of hydrochloric acid and the coloration time were investigated by univariate analyses,the results suggested optimum conditions should be as follows:the dose for both chromogenic agent and SDS was 2.0 mL,the concentration of hydrochloric acid was 0.1 mol/L,with the coloration time was 20 min.Applying above-mentioned optimized condition,the content of total phenolic acid in the extract of Taraxacum mongolicum was 120.5 mg/g.This method was simple,stable and reliable,having a good linearrelation in the range of 2.0 μg/mL-20.0 μg/mL(r=0.999 2)with the average recovery of 98.86%and RSD of 0.84%.which is suitable for analysis of total polyphenols.
