中文摘要: 对石榴籽油总多酚含量测定方法进行优化。考察提取溶剂、溶剂浓度和提取次数对石榴籽油总多酚提取效率的影响,同时以石榴籽中所含的3种多酚及油多酚提取液为研究对象,对测定方法的检测波长、Folin-Ciocalteu试剂用量、碳酸钠溶液用量、试剂加入的间隔时间和反应时间进行了优化。结果显示:石榴籽油经80%乙醇提取3次后多酚可提取完全,优于常用的甲醇提取或经正己烷脱脂处理后的甲醇提取法。以没食子酸为对照,优化后的显色条件为:检测波长765 nm,Folin试剂3.5 mL,7.5%的碳酸钠溶液2.5 mL,试剂加入间隔时间5 min,显色反应30 min。此条件下没食子酸的高、中、低浓度平均回收率为(100.69±0.34)%;没食子酸浓度在10 μg/mL~60 μg/mL范围内,吸光度与浓度线性关系良好(r=0.999 1);同一样品重复测定6次的RSD为0.67%。

Abstract:The method for the determination of total polyphenols in pomegranate seed oil was optimized in this study.The effects of extraction solvents,solvent concentration and extraction times on the extract efficiency of total polyphenols from pomegranate seed oil were examined.Meanwhile,the assay conditions,including detection wavelength,the amounts of Folin-Ciocalteu reagent and sodium carbonate solution,the interval time for the reagent addition,and the reaction time,were screened for three polyphenols in pomegranate seeds and the phenol extract from the oil,respectively.The results showed that total polyphenols in the oil was recovered completely by being extracted with 80%ethanol for three times.The extract efficiency was higher than the commonly used methods,either extraction with methanol or being defatted with hexane followed by methanol extraction.Based on the control of gallic acid,the chromogenic conditions were optimized as followed:detection at 765 nm,3.5mLofFolin-Ciocalteureagent,2.5mLof7.5%sodiumcarbonatesolution,5minofintervaltimeforthereagent addition,30 min of reaction time at 25℃.Under such conditions,the mean recovery of gallic acid at low,medium,and high concentrations was(100.69±0.34)%.The absorbance was linear with the gallic acid concentration intherangeof10μg/mLto60μg/mL(r=0.9991).TheRSDofsix-timeassaysforthesamesamplewas0.67%.
