中文摘要: 研究甜菊糖的提取工艺,以絮凝阶段为主要研究点,探讨新的絮凝剂-壳聚糖对甜菊糖浸提液絮凝工艺效果进行试验研究。在单因素试验的基础上,利用响应面法对甜菊糖浸提液的絮凝条件进行优化,建立了二次回归模型,通过分析得出,确定壳聚糖的添加工艺为:第1、4次浸提液中添加分子量为150 kDa、0.3 mL 1%壳聚糖溶液中添加1 g甜菊叶(干重)、絮凝温度为48℃、絮凝时间为2.9 h,甜菊糖的保留率为92.36%,溶液澄清率为94.17%。第2、3次浸提液中添加分子量为50 kDa的壳聚糖,添加量为0.3 mL1%壳聚糖溶液/g甜菊叶,31℃下絮凝1.9 h,甜菊糖的保留率为93.61%,溶液澄清率为96.01%。新的絮凝剂-壳聚糖避免了对生产设备的腐蚀,大大降低了生产成本,同时也减轻了对环境的污染。

Abstract:To optimize the flocculation of technology of water-extraction solution from stevia,the single factor tests and response surface methodology(RSM)were applied.Over all research was preceded by rational design.Technological condition parameters were determined according to regression analysis by SAS.The RSM was feasible for optimum flocculation process and this will improve flocculation effect.The conditions were as follows when chitosans were flocculating:added 0.3 mL/g chitosans whose molecular weight was 150 kDa into the first and the fourth extracts.The temperature was 48℃and the time was 2.9 h.Clarification rate and retention rate of the first and the fourth extracts was 94.17%and 92.36%.Added 0.3 mL/g chitosans whose molecular weight was 50 kDa into the second and the third extracts.The temperature was 31℃and the time was 1.9 h when chitosans were flocculating.Clarification rate and retention rate of the second and the third extracts was 96.01%and 93.61%.Using chitosan greatly reduces the production cost and the pollution to the environment.
