中文摘要: 对新鲜仙人掌提取多糖最优工艺及其抗氧化效果进行研究。在单因素试验的基础上采用响应面法RSM对热水浸提新鲜仙人掌多糖提取条件进行优化,优化后的工艺为:温度64.25℃,时间1.5 h,液料比22.38∶1(mL/g),提取率高达(0.114 2±0.027 3)%,多糖纯度为(37.686±9.009)μg/mL。利用体外实验模型评价其抗氧化性,结果表明该提取液对3种自由基有一定的清除能力,且呈剂量相关性,浓度分别为5.58、10.04、8.78 mg/mL的提取液可清除50%的DPPH自由基、羟自由基和ABTS+自由基。
Abstract:The extraction conditions ofO.dilleniipolysaccharide with a series of methods were examined.On the basis of one factor tests,the response surface methodology was used to optimize the boiling water extraction ofO.dilleniipolysaccharides.Design expert analysis indicated that the optimal processing conditions were:temperature 64.25 ℃,time 1.5 h,liquid to solid ratio 22.38 ∶1(mL/g),under these conditions,the extraction yield ofO.dilleniipolysaccharides was (0.114 2±0.027 3)%and mass concentration was (37.686±9.009)μg/mL.Vitro experimental model was used to evaluate the antioxidant activities,the results showed that the extracting liquid of theO.dilleniihad the capacity to scavenge three kind of free radical and scavenging activity was increased with the polysaccharide concentration.The extracting liquor with 5.58,10.04,8.78 mg/mL concentration of extracting liquor ofO.dilleniicould eliminate 50%DPPH free radical,hydroxyl radicals and ABTS+free radical.
