中文摘要: 蛋白酶 K 是一种高活性的丝氨酸蛋白酶,不但广泛用于分子生物学和细胞生物学等领域的相关实验,而且在 食品、农业、医药等领域的应用也与日俱增。从本质上说,蛋白酶 K 降解影响核酸提取的组蛋白、核酸酶等蛋白质,是其 用于各个领域的主要基础。目前,我国尚无针对核酸提取用蛋白酶 K 产品的统一标准,从而给蛋白酶 K 产品的流通、使 用和监管造成诸多不便和困难。对蛋白酶 K 的性质和酶活力测定方法进行分析,并对其在从各种生物材料提取核酸中 的应用条件进行总结,以期为核酸提取用蛋白酶 K 制定相关的方法标准和产品标准提供参考。
Abstract:Proteinase K, a kind of serine proteinases with high activity, is not only widely used in the related experiments of molecular biology and cell biology, but also increasingly applied to food, agriculture, medicine and other fields. In essence, proteinase K can degrade histone, nuclease and other proteins that affect the ex- traction of nucleic acid, which is the main foundation of its application in various fields. At present, there is not any uniform standards about proteinase K production specifically used for the extraction of nucleic acid in Chi- na, which causes much inconvenience and difficulty in the circulation, application and supervision. In this es- say, the property and detection method about enzyme activity of proteinase K were analyzed, and its actual ap- plication conditions in the extraction of nucleic acid from different biomaterials were also summarized. The paper was aimed at providing some reference for the formulation of method standards and production standards related to proteinase K used for the extraction of nucleic acid.
