中文摘要: 采用 AB-8 大孔径树脂柱层析法对龙眼壳黄酮进行纯化,对上样量、pH 值、洗脱液浓度等洗脱条件进行优化, 得最佳纯化工艺条件为:上样量为 10 mL 时,样品浓度为 0.264 mg/mL;pH 值为 3;洗脱液为 90 %的乙醇。同时对龙眼壳 黄酮 HPLC 分析条件进行优化,结果表明最优分析条件为:流动相,甲醇-水(25 ∶ 75,体积比);流速,0.5 mL/min;波长: 254 nm;柱温:25 ℃;进样量:4 μL;在此条件下经柱层析法有效纯化的龙眼壳黄酮样品中芦丁纯度最高可达 63.95 %。

Abstract:The longan shell flavonoids were purified by AB-8 macroporous resin column chromatography method, those elution conditions, such as volume of sample, pH value and elution density were optimized, and the best puri- fication process conditions were the volume of sample,10 mL; the concentration of eluent, 0.264 mg/mL; pH value, 3; eluent, 90 % ethanol. Under these HPLC conditions [C18 column, mobile phase: methanol and water (25 ∶ 75, volume ratio), flow rate: 0.5 mL/min, wavelength: 254 nm, column temperature: 25 ℃, sample quantity:4 μL], purification sample was tested and the result contrasted with rutin standard showed that the rutin concentration was up to 63.95 % in sample under the optimal process condition and the column chromatography method.
