3种食用菌分级多糖消脂护肝功能研究

中文摘要: 通过热水提取、乙醇沉淀的方式获得榆黄菇、毛木耳和灰树花多糖,利用分子量截留的超滤方法获得 15 个分 级多糖组分。采用酒精和脂肪酸(FFA)共同诱导 HepG2 细胞建立脂肪肝模型,以细胞存活率和胞内甘油三酯含量为检 测指标评价分级多糖消脂护肝作用。研究结果发现 PS V、AP V 和 GF V 3 个分子量高于 100 kDa 的组分是各自品种分级 多糖中的优势组分, 比例分别为 71.06 %、34 %和 68.89 %;3 种分级多糖可以提高肝损伤细胞的存活率分别为 27.61 %、37.68 %和 29.25 %,同时减少甘油三酯在细胞内的堆积,具有明显的消脂护肝作用。

Abstract:This study obtained crude polysaccharides of Pleurotus citrinipileatus, Auricularia polytricha and Grifola frondosabythe hot-water-extraction and ethanol-precipitation method. The crude polysaccharides were divided into 15 fractional components using ultrafiltration method of molecular weight. Fatty liver injury model was induced by alcohol and fatty acids with HepG2 cells. The cell survival rate and intracellular triglyceride levels were detected to evaluate the hepato-protective effect of fractional polysaccharides. The results showed that the components PS V, AP V and GF V, molecular weight above 100 kDa, were the highest content frac- tions each strains with the ratios of 71.06 %, 34 % and 68.89 %, respectively. PS V, AP V and GF V could in- crease the survival rate of injury hepatic cells, which were 27.61 %, 37.68 % and 29.25 %, respectively. Mean- while these fractional polysaccharides also reduced intracellular triglycerides accumulation, which indicated that they were potential agents in hepatic protection application.
