中文摘要: 研究枣仁颗粒的镇静催眠作用及其作用机制。连续 7 d 不同剂量的枣仁颗粒灌胃(10、20、40 mg/kg,以地西泮作 为阳性对照,生理盐水作为空白对照,比较各组间小鼠自主活动次数,阈下剂量戊巴比妥钠小鼠睡眠只数,阈上剂量戊 巴比妥钠小鼠睡眠潜伏期及睡眠时间,并比较小鼠脑组织中 γ-氨基丁酸(GABA)含量。结果表明,中高剂量的枣仁颗粒 均可显著减少小鼠自主活动次数,缩短活动时间(P<0.05),中高剂量枣仁颗粒可显著提高阈下剂量戊巴比妥钠小鼠的 睡眠只数(P<0.05),低中高剂量的枣仁颗粒均可缩短阈上剂量小鼠的睡眠潜伏期,延长睡眠时间(P<0.05),小鼠脑组织 中 GABA 含量升高。枣仁颗粒具有良好的镇静催眠作用,其作用机制可能为通过调节 GABA 含量发挥镇静催眠作用。

Abstract:To study the sedative and hypnotic effects of Zizyphi spinosae semen Granule and the potential mechanism. Administrated with Zizyphi spinosae semen Granule (10、20、40 mg/kg) for 7 consecutive days,di- azepam as a positive control, saline as control, The number of spontaneous activities in mice, the number of sleep deprivation in pentobarbital sodium mice, the sleep latency and the sleep time in pentobarbital sodium mice were compared, and the content of γ-amino butyric acid (GABA) in brain tissue of mice was compared. The results showed that Zizyphi spinosae semen particle moderate and high doses could significantly reduce the number of independent activity in mice, shortening the activity time(P<0.05), Zizyphi spinosae semen particle moderate and high doses could significantly raise the subthreshold dose of pentobarbital sodium in mice sleeping only a few(P<0.05), low,moderate and high doses of Zizyphi spinosae semen particle could shorten sleep laten- cy threshold dosages in mice, prolonged sleep time(P<0.05), GABA in the brain tissue of mice was significant- ly increase. Zizyphi spinosae semen particle have a good sedative and hypnotic effects, the mechanism of action may be to play a sedative and hypnotic effect by adjusting the content of GABA.
