中文摘要: 研究碘离子对色氨酸的荧光猝灭效应,旨在建立色氨酸荧光猝灭法测定海带中碘离子。在 pH=6.0 的磷酸缓冲 液中,加入不同浓度的碘离子,测定色氨酸溶液的荧光光谱及强度的变化。试验表明,色氨酸具有释放荧光的特性,最佳 激发和发射波长分别为 223 nm 和 352 nm。碘离子对色氨酸的荧光具有猝灭作用。色氨酸浓度为 10 μmol/L 时,碘离子对 色氨酸的荧光猝灭呈线性关系,根据猝灭程度可计算出碘离子的浓度,该方法的检测限为 1.64 μmol/L。根据该方法检测 了两种海带样品中的碘离子,其浓度为 637 μg/g 干重(Laminaria japonica Aresch)和 621 μg/g 干重(Saccharina japonica)。

Abstract:The novelmethod of determining iodide ion content in kelp was established by the reaction of iodide ionquenchingtryptophan fluorescenceintensity. Under phosphate buffer medium at pH = 6.0,adding different concentrations of iodide into fluorophore solutions with given concentrations,fluorescence intensives were mea- sured,tried several type of fluorophores, found tryptophan hadcharacteristics of fluorescence releasing. Experi- ments showedthat the fluorescence optimum excitation spectra of tryptophan and its emission spectra were 223 nm and 352 nm. When the concentration of tryptophan was 10 μmol/L, the iodide ionhad a liner relation of tryp- tophan fluorescence quenching. According to the degree of quenching couldcalculate the concentration of the io- dide ion,the detection limit was 1.64 μmol/L. Two kinds of kelp was tested according to the method. In Laminaria japonica Aresch the concentration of the iodide ion was 637 μg/g, and it was 621 μg/g in Saccharina japonica.
