中文摘要: 建立苯酚-硫酸和蒽酮-硫酸显色测定固态发酵红曲中可溶性多糖含量的方法, 并进行两种方法的比较研究。 固态发酵红曲采用水提取红曲多糖,所得多糖提取液用淀粉酶去除淀粉、Sevag 试剂去除蛋白质,并经醇沉得到红曲多 糖,再分别考察苯酚-硫酸和蒽酮-硫酸法两种多糖含量测定方法的线性关系、重复性、稳定性、精密度、加样回收率等指 标。苯酚-硫酸法在 30 μg~70 μg 范围内线性关系良好(R2 =0.998 1),蒽酮-硫酸法在 40 μg~120 μg 范围内线性关系良好(R2 =0.999 7)。两种方法学考察结果均较好,且测定红曲多糖的含量苯酚-硫酸法为 2.51 %,蒽酮-硫酸法为 2.52 %,两种 方法结果相近。苯酚-硫酸法和蒽酮-硫酸法均能用于测定固态发酵红曲多糖含量。
Abstract:The establishment of quantity determine method of polysaccharide of red yeast rice by phenol- sulfu- ric acid method and anthrone-sulfuric acid method was developed, and then two methods were compared. The polysaccharide was obtained with the water extraction from red yeast rice. The starch was removed from polysac- charide extracts by amylase, while the protein removed by Sevag reagent. Then the polysaccharide was obtained by ethanol precipitation. And then the study was taking on the linearity, repeatability, stability, precision and recovery index of phenol-sulfuric acid method and anthrone-sulfuric acid method. The linearity of the phenol-sulphuric acid method(R2=0.998 1)was 30 μg-70 μg, and the anthrone-sulphuric acid method(R2=0.999 7) was 40 μg-120 μg.The results of the two methods were good, and the determination of the polysaccharide con- tent of red yeast rice by the two methods was 2.51 % and 2.52 %. The results of the two methods were similar. The phenol-sulfuric acid method and anthrone-sulfuric acid method can be used for the determination of the polysaccharide content of red yeast rice.
