中文摘要: 采用醇提法研究微波协同表面活性剂提取黄芪茎总黄酮的工艺条件,并探索黄芪茎总黄酮提取液的体外抗 氧化能力。以总黄酮提取率为考察指标,在单因素试验基础上,通过正交试验优化黄芪茎总黄酮的提取条件;以黄芪 茎总黄酮提取液的 DPPH·清除率、·OH 清除率来评价其体外抗氧化活性。 试验结果表明最佳提取工艺为以 45 %乙 醇水溶液为提取溶剂,料液比为 1 ∶ 17(g/mL),微波功率为 600 W,加入药材干重 0.4 %的十六烷基三甲基溴化铵,提 取温度为 50 ℃,提取时间为 10 min。在上述提取条件下,黄芪茎总黄酮的提取率为 37.62 mg/g,该工艺比仅用微波法 提取黄芪茎总黄酮的提取率提高了 11.87 mg/g,试验结果表明,微波协同表面活性剂法提取黄芪茎总黄酮的提取率 较高,而且黄芪茎总黄酮提取液具有一定的体外抗氧化活性。
Abstract:Using ethanol as extracting agent, the extraction technology of total flavonoids (TP) in astragalus stems with surfactants assisted with microwave method and antioxidant ability of TP extraction solution in vitro were studied. Applying extraction efficiency of TP to evaluation indicator, the extraction technology of TP in as- tragalus stems was optimized on the base of the single factor experimental results. The antioxidant ability of TP extraction solution in vitro was evaluated according to its clearance ratio to both DPPH·and·OH free radical. The optimum extracting agent, ratio between material and solvent, the microwave power, the addition of CTAB (cetyl trimethyl ammonium bromide) surfactant, extracting temperature and extracting time were 45 % ethanol aqueous solution, 1 ∶ 17 (g/mL), 600 W, 0.4 % dry weight of raw medicinal materials, 50 ℃ and 10 min re- spectively. The extraction efficiency of TP could reach 37.62 mg/g under the optimum extracting condition, which increase extraction efficiency by 11.87 mg/g compared with single microwave extracting method. The ex- perimental results had shown that extraction technology of TP with surfactants assisted with microwave method was high efficient and TP extraction solution had certain antioxidant ability in vitro.
