低温低压法提取桑黄菌丝体活性多糖

中文摘要: 首次通过旋转蒸发仪在低温低压条件下提取桑黄菌丝体活性多糖, 并利用响应面法对提取条件进行了优化。 在单因素试验结果的基础上,分别以多糖得率和其总抗氧化活性为响应值,使用 Minitab 16 软件对提取时间、压力、液 料比进行了优化,并通过响应面优化器对同时获得高活性高产率桑黄多糖的条件进行了优化。结果显示,在提取时间、 压力和液料比分别为 1.8 h、0.059 MPa 和 52.5 ∶ 1(mL/g)的最优提取条件下,从桑黄菌丝体中获得多糖的得率和抗氧化 活性(TEAC)分别可达到 4.41 %和 6.33 mM。证实低温低压法提取真菌活性多糖的方法是可行的。
Abstract:The crude polysaccharide with antioxidant activity was firstly extracted from mycelia of Phellinus sp. using a rotary evaporation apparatus under low temperature and low pressure conditions. The extraction condi- tions including extraction time, pressure and solvent -solid ratio were optimized through response surface method(RSM), in which the individual yield and total antioxidant activity of polysaccharide were chosen as re- sponse values. Based on the results of RSM experiments, the response optimizer was further used to simultane- ously get high yield and high activity of polysaccharide during extraction. Results showed that the optimum ex- traction conditions were extraction time 1.8 h, pressure 0.059 MPa, solvent-solid ratio 52.5 ∶ 1 (mL/g), re- spectively. The polysaccharide yield and Trolox-Equivalent Antioxidant Capacity(TEAC) reached 4.41 % and 6.33 mM, respectively, under optimum extraction conditions. A new feasible extracting method for crude polysaccharide from mycelia of Phellinus sp. with high yield and high total antioxidant activity has been devel- oped just using a rotary evaporation apparatus.
