微波辅助萃取马兰头抑制黑曲霉活性成分的工艺优化

中文摘要: 以马兰头(Kalimeris indica L.)为原料,在单因素试验的基础上,利用正交试验法优化微波辅助萃取马兰头抑制 黑曲霉活性成分的工艺。结果表明,各因素对萃取马兰头抑制黑曲霉活性成分影响程度大小顺序为:料液比>温度>功 率>时间,并得到最佳萃取工艺为功率 700 W,温度 70 ℃,时间 3 min,料液比 1 ∶ 70(g/mL),验证表明试验结果真实可信。 采用最佳萃取工艺制得马兰头抑制黑曲霉活性成分粗品的产率为 44.62 %, 进一步试验结果表明该提取物对黑曲霉(Aspergillus niger)、青霉(Penicillium)均有较好的抑制效果,其对黑曲霉、青霉的 MIC 均约为 0.94 mg/mL,表明马兰头提 取物具有抑制真菌生长的潜能。
Abstract:On the basis of single factor experiment, the microwave-assisted extraction technique of bioactive compounds of Aspergillus niger inhibitors from Kalimeris indica L. was optimized by orthogonal design. The re- sults showed that the primary and secondary order of influence of the factors microwave -assisted extraction method for bioactive compounds of Aspergillus niger inhibitors from Kalimeris indica L. was confirmed: the ratio of solid to liquid>temperature >power >time. The optimum process condition was confirmed that the yields of crude extract was 44.62 % could be obtained when extraction power was 700 W, extraction temperature was 70 ℃, extraction time was 3 min, the ratio of solid to liquid was 1 ∶ 70 (g/mL). Verification showed the result was authentic. Further experimental results showed that the extract had better inhibitory activity on Aspergillus niger and Penicillium. The Minimum Inhibition Concentration (MIC) for Aspergillus niger and Penicillium are both about 0.94 mg/mL, showed the extract of Kalimeris indica L. has the potential to inhibit fungal growth.
