中文摘要: 研究高渗环境中假丝酵母(T 酵母)细胞内海藻糖和甘油的代谢情况。利用微波破碎法这一传统的物理破壁法 来提取 T 酵母细胞内甘油和海藻糖,并通过高效液相色谱仪测定细胞内甘油和海藻糖的含量。经过高效液相色谱测定, 可以得到不同盐浓度下 T 酵母胞内甘油和海藻糖含量。T 酵母在高渗环境中通过对甘油和海藻糖的积累,同时阻止甘 油的外排作用来抵抗外界环境对其生长的影响。

Abstract:To study the metabolism of trehalose and glycerol in the cells of Candida Versatilis (T yeast) in the high osmotic environment. Using microwave crushing method the traditional physical wall breaking method to extract t yeast intracellular glycerol and trehalose,and by high performance liquid chromatography for the deter鄄 mination of the content of intracellular glycerol and trehalose. The contents of glycerol and trehalose in T yeast cells under different salt concentrations were determined by HPLC. The accumulation of glycerol and trehalose in the hypertonic environment of T yeast was also prevented by blocking the efflux of glycerol.
