中文摘要: 探究绿色木霉自絮凝固定化条件,以便提高其分泌纤维素酶的能力。首先测定固定化时间、体积、pH、葡萄糖与 蛋白胨浓度对自絮凝固定化绿色木霉分泌纤维素酶的影响,然后用正交试验设计进行优化组合。时间、体积和 pH 对自 絮凝绿色木霉分泌纤维素酶的影响均呈钟形曲线;葡萄糖对自絮凝绿色木霉分泌纤维素酶影响不大;蛋白胨对自絮凝 绿色木霉分泌纤维素酶影响较大。绿色木霉自絮凝固定化最优条件是:时间为 6 d,pH 为 6,体积为 70 mL,葡萄糖为2.25 %,蛋白胨为 1.50 %。按此条件培养自絮凝固定化的绿色木霉 72 h,分泌的纤维素酶活性为 3.55 U。
Abstract:Researching on the conditions of Trichoderma viride immobilized with self-flocculation,so we could get high cellulase activity. First,we measured the effects of time,volume,pH,glucose and peptone concentra鄄 tions on cellulase activity of self-flocculating immobilized Trichoderma viride. Then we used the orthogonal test design to obtain optimized combinations. The times,volumes and pH had a bell shaped curve effect on cellulase activity of immobilized Trichoderma viride with self-flocculating. The glucoses had small effect on cellulase ac鄄 tivity,but the peptone had great effect on cellulase activity. The best optimal conditions of self-flocculation of immobilized Trichoderma viride were 6 d of time,70 mL of volume,6 of pH,2.25 % of glucoses and 1.50 % of peptones. Under the optimized conditions,the cellulase activity could reach 3.55 U after Trichoderma viride for cultivating of 72 h.
