中文摘要: 以还原型谷胱甘肽和抗坏血酸为对照, 采用 DPPH·法和邻二氮菲-Fe2+氧化法分别测定鱼皮低聚肽清除 DPPH·和·OH 活性能力;并利用 GB/T 5009.124-2003《食品中氨基酸的测定》方法测定鱼皮低聚肽的氨基酸组成,进行营 养价值评价及抗氧化能力与氨基酸组成关系的初步分析。结果表明,在优选的反应体系中,鱼皮低聚肽、谷胱甘肽和抗坏血 酸对·OH 的 IC50 值分别为 2.482、0.357、0.100 mg/mL;对 DPPH·的 IC50 值分别为 0.635、0.030、0.003 mg/mL;鱼皮低聚肽必 需氨基酸含量为 24.43 %,EAA/TAA 值为 24.19 %,EAA/NEAA 值为 32 %,CS 均大于 100,EAAI 为 352;含硫氨基酸含量 为 3.8 %,除蛋氨酸外的疏水性氨基酸占氨基酸总量的 26.7 %,具有抗氧化活性的氨基酸总量为 30.5 %,与鱼皮低聚肽 的抗氧化活性一致。
Abstract:Under the optimum conditions using glutathione and ascorbic acid as the controls, DPPH· and phenanthroline-Fe2+ oxidation assays were applied to evaluate the capacities for scavenging free radicals of fish skin oligopeptides. Determination of amino acid composition of fish skin oligopeptides put to use the GB/T 5009.124-2003 (Determination of amino acids in foods) method. The relationship between antioxidative activity and amino acid composition of fish skin oligopeptides was analyzed, and the nutritional values of those were evaluated. The results showed that the values of half-inhibitory concentration (IC50) of fish skin oligopeptides, glutathione and ascorbic acid for·OH were 2.482, 0.357, 0.100 mg/mL, respectively, and those for DPPH· were 0.635, 0.030 and 0.003 mg/mL, respectively. Essential amino acids contents of fish skin oligopeptides were 24.43 %, EAA/TAA was 24.19 %, EAA/NEAA was 32 %, CS was greater than 100, and EAAI was 352. Sulphur amino acids contents were 3.8 %, hydrophobic amino acids except methionine accounted for 26.7 % of total amino acids, so the total antioxidative activity amino acids were 30.5 % which was consistent with the antioxidative activity of fish skin oligopetides.
