滁菊水提物抑制高脂饮食诱导的氧化应激

中文摘要: 探讨滁菊水提物对高脂饮食诱导的内脏沉积及氧化应激的影响。将 3 周龄 ICR 小鼠[(10±2)g]随机分为 3 组, 每组 7 只:对照组(饲喂基础饲料),高脂饮食组(饲喂高脂饲料)、滁菊干预组(饲喂高脂饲料+饮用滁菊水提物)。饲养 8周后测量体重,处死后取附睾、肾周脂肪垫称重计算脂体比,收集血清样本并检测血清丙二醛(MDA)、总谷胱甘肽 (GSH)水平及超氧化物岐化酶(SOD)、过氧化氢酶(CAT)酶活性。与对照组相比,高脂组脂体比、血清 MDA 水平显著增 高,而 GSH 水平及 SOD、CAT 活性则显著降低。与高脂组相比,滁菊干预组脂体比、血清 MDA 水平均显著降低,而 GSH 水平及 SOD、CAT 活性则显著升高。结果表明,高脂饮食可诱导内脏脂肪沉积及氧化应激,滁菊干预可通过保护抗氧化 系统抑制高脂饮食诱发的以上危害。

Abstract:To explore the effects of aqueous extract from chrysanthemum morifolium of Chuju on visceral fat accumulation and oxidative stress induced by high-fat diet. Three-week old ICR mouse weighing [(10±2) g] were divided into three groups, seven for each group; groupⅠ:received normal diet, groupⅡ:received high-fat diet, and groupⅢ: cotreated with high-fat diet and Chuju aqueous extract. All animals were fed for 8 weeks, and measured the body weight at the end, then separated the visceral fat pad and weighed it after sacrificing mouse to calculate the ratio of fat to body mass. Blood samples were taken for measurement of serum levels of MDA, GSH and the enzymatic activities of SOD, CAT. Compared by control, the high-fat diet group showed larger ratio of weight of visceral fat to body, higher serum MDA level, but lower level of GSH and reduced enzymatic activities of SOD and CAT; however, compared with the latter, the above effects of high-fat diet were reversed by exposure to aqueous extract of Chuju. Results described above suggest that high-fat diet could trigger visceral fat accumulation and oxidative stress which being inhibited with co-treatment of Chuju aqueous extract.
