中文摘要: 为有效利用罗非鱼皮制备具有抗氧化活性的胶原三肽,该研究采用响应面法优化酶解条件制备罗非鱼皮胶原三肽,并通过对比其与市售胶原三肽的分子量分布、氨基酸组成、抗氧化活性的差异,评价罗非鱼皮胶原三肽的品质。结果表明,利用碱性蛋白酶和菠萝蛋白酶复配酶解(加酶量为5 000U/g,酶活比为1∶1),在酶解温度为55 ℃、液料比为6.3∶1(mL/g)、酶解pH值为9.2的条件下,制备的罗非鱼皮胶原三肽的含量最高,为37.25%。与市售胶原三肽相比,罗非鱼皮胶原三肽具有更小的重均分子量(476 Da),更高含量的必需氨基酸(15.42%)和疏水性氨基酸(32.53%)。此外,当罗非鱼皮胶原三肽浓度为2.00 mg/mL时,DPPH自由基和ABTS+自由基清除率分别达38.55%和61.97%,表现出优于市售胶原三肽的抗氧化能力。
Abstract:To effectively utilize tilapia skin for the preparation of collagen tripeptides with antioxidant activity,response surface methodology was adopted to optimize the enzymatic hydrolysis conditions. The quality of tilapia skin's collagen tripeptide was evaluated by comparing its molecular weight distribution,amino acid composition,and antioxidant activity with those of commercial collagen tripeptide. The results showed that the highest tripeptide content of 37.25% was obtained using a compound enzyme of alkaline protease and bromelain (enzyme dosage od 5 000 U/g at a ratio of 1∶1) under the conditions of enzymatic hydrolysis temperature of 55 ℃,liquid-solid ratio of 6.3∶1(mL/g),and pH of 9.2. Compared with commercial collagen tripeptide,the tilapia skin's collagen tripeptide had a smaller weight-average molecular weight (476 Da) and higher contents of essential amino acids (15.42%) and hydrophobic amino acids (32.53%). Furthermore,at a concentration of 2.00 mg/mL,the DPPH and ABTS+free radical scavenging rates of tilapia skin's collagen tripeptide reached 38.55% and 61.97%,respectively,indicating superior antioxidant capacity compared to commercial collagen tripeptide.
