中文摘要: 该研究以一株产β-葡萄糖苷酶鸡乳杆菌FB20发酵胡萝卜渣提取水溶性膳食纤维(soluble dietary fiber,SDF)和不溶性膳食纤维(insoluble dietary fiber,IDF),在基于细胞和斑马鱼胚胎毒性试验评价其安全性的基础上,通过高效液相色谱、扫描电子显微镜(scanning electron microscope,SEM)对两种膳食纤维结构进行分析,通过测定促益生菌生长能力、体外模拟消化情况、体外发酵后微生物群落和短链脂肪酸含量的变化,对两种膳食纤维作为益生元的功能进行评价。结果显示,所提取的两种膳食纤维均具有安全性,两者单糖组成和微观结构具有一定差别;在功能方面,两种膳食纤维均具有对植物乳植杆菌FB13、短双歧杆菌FB18和鼠李糖乳酪杆菌GG的生长促进作用,且IDF组相较于SDF组效果更优;在模拟消化液中二者均不会被消化,体外发酵试验显示SDF组相较于空白组和IDF组在一定程度上可以促进肠道微生物的平衡,拟杆菌门和厚壁菌门中部分有益菌的菌群丰度得到提升,丁酸、乙酸、己酸和戊酸等短链脂肪酸的含量明显更高。
Abstract:This study employed Lactobacillus gallinarum FB20,a β-glucosidase-producing strain,to ferment carrot pomace for the extraction of soluble dietary fiber (SDF) and insoluble dietary fiber (IDF). The safety of both fiber types was confirmed through cell-based assays and zebrafish embryo toxicity tests. Structural and prebiotic properties were characterized by analyzing monosaccharide composition and scanning electron microscopy,alongside evaluations of probiotic growth promotion,in vitro simulated digestion,and changes in microbial composition and short-chain fatty acid (SCFA) profiles after in vitro fermentation. The results indicated that both types of dietary fibers are safe for consumption. Structural analyses revealed distinct monosaccharide compositions and microstructural features between SDF and IDF. Both fibers enhanced the growth of Lactiplantibacillus plantarum FB13,Bifidobacterium breve FB18,and Lactobacillus rhamnosus GG,with IDF demonstrating superior performance compared to SDF. During in vitro simulated digestion,neither fiber was degraded in the oral or gastric phases. In vitro fermentation results showed that,compared to the control and IDF groups,the SDF group promoted a more balanced gut microbiota composition,notably increasing the relative abundances of the benefit bacteria in the phylum of Bacteroides and Firmicutes,etc. Additionally,the concentrations of SCFAs—including butyric,acetic,hexanoic,and pentanoic acids—were significantly higher in the SDF group.
