中文摘要: 为提高山苏的利用价值,该试验采用超声辅助酶解提取工艺提取山苏粉,以蛋白质含量为指标,通过单因素试验分析超声频率、功率密度、超声时间和酶解条件等因素对山苏提取效果的影响,并探究其对2型糖尿病小鼠的降血糖、降血脂及降尿酸功效。结果表明,最佳工艺条件为超声频率20 kHz、功率密度1 950 W/L、超声时间10 min,并使用纤维素酶和果胶酶进行酶解,酶质量比为1∶1、酶浓度为20 U/mL、pH4.5、酶解温度50 ℃、酶解时间60 min。该工艺下山苏粉的蛋白质含量和可溶性膳食纤维含量提高。与模型对照组相比,高剂量组(500 mg/kg)使糖尿病小鼠空腹血糖降低49.1%,总胆固醇、甘油三酯和低密度脂蛋白胆固醇水平分别降低了55.8%、21.6%和28.0%,尿酸水平降低了33.4%。
Abstract:To improve the utilization value of Asplenium nidus,this study adopted an ultrasonic-assisted method to prepare A. nidus powder. Taking protein content as the evaluation index,single-factor experiments were carried out to analyze the effects of ultrasonic frequency,power density,ultrasonic time and enzymatic hydrolysis conditions on the extraction efficiency of A. nidus. Meanwhile,the hypoglycemic,hypolipidemic and uric acid-lowering activities of the obtained powder were investigated in type 2 diabetic mice. The results showed that the optimal technological conditions were determined as follows:ultrasonic frequency of 20 kHz,power density of 1 950 W/L,ultrasonic time of 10 min,compound enzymolysis with cellulase and pectinase at the mass ratio of 1∶1,enzyme concentration of 20 U/mL,pH value of 4.5,enzymolysis temperature of 50 ℃,and enzymolysis time of 60 min. Under the optimized conditions,the protein content and soluble dietary fiber content of A. nidus powder were effectively increased. Compared with the model control group,the high dose of A. nidus powder (500 mg/kg) reduced the fasting blood glucose of diabetic mice by 49.1%. The levels of total cholesterol,triglyceride and low-density lipoprotein cholesterol were decreased by 55.8%,21.6% and 28.0%,respectively,and the serum uric acid level was reduced by 33.4%.
