中文摘要: 为开发安全高效的带鱼冷链保鲜技术,评价不同工艺制备的植物乳植杆菌(Lacticaseibacillus plantarum)SC019后生元的抑菌活性及保鲜效果,该文分别采用热灭活(heat-inactivated,HI)与超声破碎(ultrasonic,US)制备后生元(HI-/US-POST SC019),以无菌水作为对照,测定其抑菌圈直径、生物膜抑制率与DNA泄漏量,并在4 ℃条件下贮藏带鱼12 d,动态监测菌落总数(total viable count,TVC)、挥发性盐基氮(total volatile basic nitrogen,TVB-N)含量、白度及感官评分。结果表明,HI-POST SC019对大肠杆菌、金黄色葡萄球菌的抑菌圈直径分别为(16.50±1.06) mm和(15.94±0.38)mm,生物膜抑制率达82.14%和80.06%,DNA泄漏量显著高于US-POST SC019(P<0.05)。贮藏试验显示,HI-POST SC019将TVB-N含量超过30 mg/100 g的时间延后至第12天,且12 d内TVC维持在6 lg(CFU/g)以下,并使感官评分始终高于可接受阈值。与对照组相比,HI-POST SC019将带鱼冷藏货架期延长约4 d。综上,热灭活工艺可显著增强SC019后生元的多靶点抑菌活性,延缓带鱼腐败进程并保持色泽与口感。
Abstract:To develop a safe and efficient cold chain preservation technology for Trichiurus lepturus,this study aimed to evaluate the antibacterial activity and preservation effect of postbiotics from Lacticaseibacillus plantarum SC019 prepared by different processes. Postbiotics were prepared via heat-inactivated (HI)and ultrasonic(US) disruption,respectively,with sterile water as the control. The inhibition zone,biofilm inhibition rate,and DNA leakage were determined. Meanwhile,Trichiurus lepturus was stored at 4 ℃ for 12 d,and the total viable count (TVC),total volatile basic nitrogen (TVB-N),whiteness,and sensory score were dynamically monitored.The results showed that the inhibition zones of HI-POST SC019 against E. coli and S. aureus were (16.50±1.06) mm and (15.94±0.38) mm,respectively,with biofilm inhibition rates reaching 82.14% and 80.06%. The DNA leakage of HI-POST SC019 was significantly higher than that of US-POST SC019 (P<0.05). Storage experiments indicated that HI-POST SC019 delayed the time when TVB-N exceeded 30 mg/100 g to the 12th day;during the 12-day storage period,TVC remained below 6 lg(CFU/g),and the sensory score was consistently higher than the acceptable threshold. Compared with the control,HI-POST SC019 extended the refrigerated shelf life of Trichiurus lepturus by approximately 4 d. Therefore,the heat inactivation process can significantly enhance the multi-target antibacterial activity of SC019 postbiotics,delay the spoilage process of Trichiurus lepturus,and maintain its color and texture.
