中文摘要: 该文以石硖龙眼核为研究对象,使用60%乙醇制备龙眼核多酚提取物,用有机溶剂进行萃取,分别得到乙酸乙酯相、二氯甲烷相、正丁醇相和萃取后水相。对龙眼核不同萃取相总酚、总黄酮、总原花青素和总单宁含量进行测定;通过脂多糖(lipopolysaccharide,LPS)诱导RAW264.7细胞建立炎症模型,检测不同萃取相龙眼核多酚对RAW264.7细胞一氧化氮(nitric oxide,NO)、活性氧(reactive oxygen species,ROS)释放量、细胞炎症因子浓度的影响;最后使用超高效液相色谱串联质谱(liquid chromatography-tandem mass spectrometry,LC-MS/MS)对多酚进行成分鉴定分析。结果表明:乙酸乙酯相总酚、总黄酮、总原花青素和总单宁含量分别为(635.56±15.55)mg GAE/g DW、(109.96±2.31)mg RE/g DW、(24.06±1.39)mg EPE/g DW、(8.22±0.05)mg TAE/g DW,在4 个萃取相中均最高;乙酸乙酯相龙眼核多酚对RAW264.7细胞NO、ROS、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)释放均有一定的抑制作用,且都具有浓度依赖性,与其余相比较整体抗炎效果较好;乙酸乙酯相中主要检出20 种酚类物质,以酚酸和黄酮类多酚为主,其中儿茶素相对含量最高。综上所述,乙酸乙酯相龙眼核多酚具有一定的抗炎潜力。
Abstract:This study focuses on 'Shixia' longan seed polyphenols. The crude polyphenol extract of longan seed was prepared with 60% ethanol,and sequential organic solvent extraction afforded four distinct phases:ethyl acetate,dichloromethane,n-butanol,and aqueous phases. The total phenolic,flavonoid,proanthocyanidin and tannin contents were determined in different extraction phases of longan seeds. An inflammatory model was established in RAW264.7 cells by lipopolysaccharide (LPS) stimulation. The effects of different extraction phases of longan seed polyphenols on nitric oxide (NO) release,reactive oxygen species (ROS) levels,and inflammatory cytokine production in RAW264.7 cells were evaluated. Finally,liquid chromatography-tandem mass spectrometry (LC-MS/MS) was employed to identify the polyphenolic components in the active extract phase. The results indicated that the ethyl acetate fraction contained the following amounts of total phenols,total flavonoids,total proanthocyanidins,and total tannins:(635.56±15.55) mg GAE/g DW,(109.96±2.31) mg RE/g DW,(24.06±1.39) mg EPE/g DW,and (8.22±0.05) mg TAE/g DW. Ethyl acetate fraction polyphenols exhibited concentration-dependent inhibitory effects on nitric oxide (NO) release,reactive oxygen species(ROS) levels,tumor necrosis factor-α (TNF-α),interleukin-6 (IL-6),and prostaglandin E2 (PGE2) in RAW264.7 cells,and its overall anti-inflammatory effect was superior compared with the other fractions. The ethyl acetate fraction contained 20 phenolic compounds,mainly phenolic acids and flavonoids,among which catechin had the highest relative content. In conclusion,ethyl acetate-extracted polyphenols from longan seeds demonstrate potential anti-inflammatory activity.
