中文摘要: 为探讨多肽与花色苷的分子互作机制及其抗氧化协同作用,采取荧光猝灭法和1,1-二苯基-2-苦基肼基自由基(1,1-diphenyl-2-picrylhydrazyl,DPPH)、2,2'-联氮-双-(3-乙基苯并噻唑啉-6-磺酸)[2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid),ABTS]和铁离子还原抗氧化能力(ferric reducing antioxidant power,FRAP)抗氧化评价体系,结合Chou-Talalay联合指数(combination index,CI)及协同系数(synergistic effect coefficient,SE),初步探索Val-Pro-Trp(VPW)与矢车菊素-3-O-葡萄糖苷(cyanidin-3-O-glucoside,C3G)分子相互作用及其在体外抗氧化能力的协同效应。结果表明:C3G可以显著猝灭VPW的荧光,呈现剂量依赖关系。在中性条件下,由猝灭常数计算结合能力为-27.28 kJ/mol,表明二者有潜在的相互作用,并通过疏水作用自发形成具有单个结合位点VPW-C3G复配物。当VPW和C3G质量浓度比为1∶1时,在DPPH、ABTS和FRAP模型中均存在协同抗氧化作用,其DPPH和ABTS+自由基清除联合作用指数分别为0.82和0.86,FRAP试验协同系数为0.05。

Abstract:This study explored the molecular interaction mechanism and synergistic antioxidant effects between the peptide and anthocyanin. The fluorescence quenching method and three in vitro antioxidant evaluation systems,including 1,1-diphenyl-2-picrylhydrazyl (DPPH),2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS),and ferric reducing antioxidant power (FRAP) assays,were adopted. Combined with the Chou-Talalay combination index (CI) and synergistic effect coefficient (SE),the molecular interaction characteristics and in vitro synergistic antioxidant effects of Val-Pro-Trp (VPW) and cyanidin-3-O-glucoside(C3G) were systematically investigated. The results showed that C3G significantly quenched the fluorescence of VPW in a dose-dependent manner. Under neutral conditions,the calculated Gibbs free energy (ΔG) was-27.28 kJ/mol,which indicated that VPW and C3G had potential intermolecular interactions and spontaneously formed VPW-C3G complexes with a single binding site through hydrophobic interaction. When the mass concentration ratio of VPW to C3G was 1∶1,significant synergistic antioxidant effects were observed in DPPH,ABTS and FRAP evaluation models. The combination indices for DPPH and ABTS+radical scavenging activities were0.82and0.86,respectively,while synergistic effect coefficient from the FRAP assay was 0.05.
