人参姜枣饮改善胃溃疡的功能因子分析及其提取工艺优化

中文摘要: 为研究人参姜枣饮改善胃溃疡的功能因子和最佳提取工艺,该试验采用液相色谱-串联质谱法分析人参姜枣饮化学成分,利用中药系统药理学数据库与分析平台数据库预测化学成分靶点,通过Pubchem数据库查询胃溃疡靶点,使用STRING和DAVID数据库分析核心靶点和相关通路,利用Cytoscape 3.10.1构建“活性成分-作用靶点-信号通路”网络,根据度值筛选人参姜枣饮功能因子,并使用分子对接技术验证核心成分和核心靶点的结合能力。最后通过单因素试验和正交试验,以功能因子总黄酮含量和提取物得率为指标优化其提取工艺。结果表明,人参姜枣饮中共鉴定出38种化合物,包括18种黄酮类、8种皂苷类、4种有机酸类、2种糖类及6种其他类化合物。网络药理学结果显示异牧荆素、异甘草素、汉黄芩素、芦丁、甘草苷等黄酮类成分为主要功能因子,作用于AKT1、TNF、MMP9、NFKB1和TP53等核心靶点,通过调控血管内皮生长因子信号通路和表皮生长因子受体信号通路等信号通路改善胃溃疡。通过正交试验确定人参姜枣饮最佳提取工艺条件为液料比120∶1(mL/g)、浸泡时间40min、提取时间60min、提取2次。在此条件下,人参姜枣饮总黄酮含量14.7 mg/g,提取物得率39.78%。
Abstract:This study aimed to explore functional components and optimal extraction parameters of ginsengginger-jujube decoction against gastric ulcer. Liquid chromatography-tandem mass spectrometry was used to identify chemical compositions of the decoction. Targets of chemical ingredients were predicted via the traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP) database,and gastric ulcer-related targets were retrieved from the PubChem database. Core targets and associated signaling pathways were analyzed using STRING and DAVID databases. A network of active component-target-signaling pathway was established with Cytoscape 3.10.1. Functional factors were screened based on degree values,and molecular docking was performed to verify binding affinity between core components and key targets. Singlefactor and orthogonal experiments were conducted to optimize extraction conditions with total flavonoid content and extraction yield as evaluation indicators.The results indicated that 38 compounds were successfully identified,consisting of 18 flavonoids,8 saponins,4 organic acids,2 saccharides and 6 other substances. Network pharmacological analysis demonstrated that flavonoids including isovitexin,isoliquiritigenin,wogonin,rutin and liquiritin served as predominant functional factors. These ingredients targeted core genes AKT1,TNF,MMP9,NFKB1 and TP53,and relieved gastric ulcer symptoms by modulating vascular endothelial growth factor signaling pathway (VEGF) and epidermal growth factor receptor signaling pathway (EGFR). Orthogonal tests confirmed the optimal extraction conditions:liquid-solid ratio 120∶1(mL/g),soaking duration 40 min,single extraction duration 60 min,and two extraction cycles. Under the optimized conditions,the total flavonoid content was 14.7 mg/g,and the extraction yield reached 39.78%.
