大鲵蛋白肽的制备及其抗氧化、抗疲劳活性

中文摘要: 人工养殖大鲵具有较高的营养和药用价值,目前产品的开发主要集中在大鲵肉制品,对其富含蛋白质的皮和黏液缺乏高值化开发利用。该研究采用碱溶酸沉法从大鲵皮和黏液中提取大鲵蛋白,利用复合蛋白酶酶解技术制备大鲵蛋白肽,通过小鼠体内外实验评估其抗氧化、抗疲劳活性。体外实验显示,大鲵蛋白肽(5 mg/mL)对超氧阴离子自由基、羟基自由基、1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基及2,2′-联氮-双-3-乙基苯并噻唑啉-6-磺酸[2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonate),ABTS]阳离子自由基清除率分别为69.09%、43.69%、61.20%和82.79%,其抗氧化活性与谷胱甘肽(glutathione,GSH)相当且呈浓度依赖性。小鼠体内实验表明,饲喂大鲵皮蛋白肽和黏液蛋白肽的小鼠肝糖原含量分别较对照组提升23%和18%。同时,两组血浆乳酸脱氢酶的活性分别达到140 U/mL和160 U/mL,高于对照组,血浆中尿素氮含量降低20.0%~22.5%,运动后血乳酸含量下降10%~14%,表明大鲵蛋白肽可以加速乳酸和尿素氮的代谢,减轻运动性肌肉损伤与疲劳。

Abstract:Artificially cultured Andrias davidianus has high nutritional and medicinal values. At present,product development mainly focuses on Andrias davidianus meat products,while there is a lack of high-value development and utilization of its protein-rich skin and mucus. In this study,Andrias davidianus protein was extracted from the skin and mucus of Andrias davidianus by alkali extraction and acid precipitation method,and Andrias davidianus protein peptides were prepared by compound protease hydrolysis technology. Their antioxidant and anti-fatigue activities were evaluated through in vitro and in vivo experiments on mice. In vitro experiments showed that the scavenging rates of Andrias davidianus protein peptides (5 mg/mL) on superoxide anion radicals,hydroxyl radicals,1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals,and 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulphonate) (ABTS) cation radicals were 69.09%,43.69%,61.20%,and 82.79%,respectively.Its antioxidant activity was comparable to that of glutathione (GSH) and was concentration-dependent. In vivo experiments on mice indicated that the liver glycogen contents of mice fed with Andrias davidianus skin protein peptides and mucus protein peptides were increased by 23% and 18%,respectively,compared with the control group. Meanwhile,the activities of plasma lactate dehydrogenase in the two groups reached 140 U/mL and 160 U/mL,respectively,which were significantly higher than those in the control group. The content of urea nitrogen in plasma was decreased by 20.0%-22.5%,and the content of blood lactic acid after exercise was decreased by 10%-14%. These results suggested that Andrias davidianus protein peptides could accelerate the metabolism of lactic acid and urea nitrogen and alleviate exercise-induced muscle damage and fatigue.
