中文摘要: 采用响应面优化复合酶辅助超声提取绞股蓝皂苷的最佳工艺并研究其体外抗氧化及降糖活性。试验结果表明,绞股蓝皂苷的最佳提取工艺条件为复合酶(果胶酶∶纤维素酶)比例2∶1 (U/U)、复合酶添加量192 U/g 底物、酶解超声时间60 min、温度63 ℃、超声功率360 W、乙醇浓度70%、液料比55∶1 (mL/g),此条件下绞股蓝皂苷提取率为9.83%。绞股蓝皂苷提取物清除DPPH自由基和ABTS+自由基的半抑制浓度(half maximal inhibitory concentration,IC50)值分别为636.14、384.05 μg/mL,对α-葡萄糖苷酶活性抑制率的IC50值为451.98 μg/mL,说明绞股蓝皂苷具有一定的体外抗氧化和降糖活性,有较好的开发应用前景。
Abstract:Response surface methodology was used to optimize the compound enzyme-assisted ultrasonic extraction of gypenosides from Gynostemma pentaphyllum and to investigate their in vitro antioxidant and hypoglycemic activities. The experimental results showed that the optimal extraction conditions for gypenosides were as follows: a compound enzyme ratio (pectinase∶cellulase) of 2∶1 (U/U), a compound enzyme dosage of 192 U/g substrate, an enzymatic hydrolysis-ultrasonic time of 60 min, a temperature of 63 ℃, an ultrasonic power of 360 W, an ethanol concentration of 70%, and a liquid-to-material ratio of 55∶1 (mL/g). Under these conditions, the extraction yield of gypenosides was 9.83%. The half-maximal inhibitory concentration (IC50)values of the gypenoside extracts for scavenging DPPH and ABTS+ radicals were 636.14 and 384.05 μg/mL,respectively, and the IC50 value for inhibition of α-glucosidase activity was 451.98 μg/mL, indicating that gypenosides possess certain in vitro antioxidant and hypoglycemic activities and have good potential for development and application.
