中文摘要: 为优化桑黄多糖提取工艺并评价其抗肺癌活性,采用热水回流法提取多糖,结合单因素与正交试验确定关键参数,构建粒子群优化-反向传播神经网络(particle swarm optimization-back propagation neural network,PSO-BPNN)模型进行工艺优化。通过网络药理学、分子对接及MTT实验探讨其抗肺癌机制。结果表明:最佳提取条件为提取温度82.5 ℃、提取时间1.75 h、料液比1∶25 (g/mL),该条件下多糖得率为6.78%;网络药理学分析筛选出73 个潜在靶点,核心靶点包括热休克蛋白90α亚型A1(heat shock protein 90α isoform A1,HSP90AA1)、苏氨酸激酶1(serine/threonine kinase 1,AKT1)、细胞肉瘤基因(cellular sarcoma gene,SRC)和激酶插入结构域受体(kinase insert domain receptor,KDR)等,主要涉及磷脂酰肌醇3激酶-蛋白激酶B信号通路(phosphatidylinositol 3-kinase/protein kinase B signaling pathway,PI3K-Akt)及癌症相关通路;分子对接显示核心成分与靶点结合能均低于-20.92 kJ/mol;MTT实验证实桑黄多糖对Lewis肺癌细胞具有浓度依赖性抑制作用,半数抑制浓度为0.31 mg/mL。
Abstract:To optimize the extraction process of Sanghuangporus vaninii polysaccharides and evaluate their antilung cancer activity,polysaccharides were extracted using a hot-water reflux method. Key parameters were determined through single-factor and orthogonal experiments,and a particle swarm optimization-back propagation neural network (PSO-BPNN) model was constructed for process optimization. The anti-lung cancer mechanisms were explored through network pharmacology,molecular docking,and MTT assays. The results showed that the optimal extraction conditions were an extraction temperature of 82.5 ℃,an extraction time of 1.75 h,and a solidto-liquid ratio of 1∶25(g/mL),with a polysaccharide yield of 6.78%. Network pharmacology analysis identified 73 potential targets,with core targets including heat shock protein 90α isoform A1 (HSP90AA1),serine/threonine kinase 1 (AKT1),cellular sarcoma gene(SRC),and kinase insert domain receptor (KDR),mainly involving the phosphatidylinositol 3-kinase/protein kinase B signaling pathway (PI3K-Akt) and cancer-related pathways.Molecular docking showed that the binding energies between the core components and the targets were all lower than -20.92 kJ/mol. MTT assays confirmed that S. vaninii polysaccharides exerted a concentration-dependent inhibitory effect on Lewis lung cancer cells,with an half maximal inhibitory concentration of 0.31 mg/mL.
