中文摘要: 为考察超声波技术对大豆分离蛋白(soybean protein isolate,SPI)生产中的豆渣残留蛋白含量和大豆乳清残留蛋白含量的影响,该试验研究了豆渣中的蛋白与SPI中的蛋白的二级结构差异,以及超声处理萃取工段对系统排出物(豆渣和大豆乳清)残留蛋白含量的影响。结果表明,与SPI相比,豆渣蛋白的二级结构中α-螺旋含量降低18.83%,β-折叠含量降低13.40%,无规则卷曲含量降低9.40%,β-转角含量升高41.60%;超声处理为5 min时,豆渣中的残留蛋白含量提高了0.4%。超声处理豆乳的大豆乳清,超声处理时间与大豆乳清残留蛋白含量之间无相关性。超声处理酸沉料液的大豆乳清,其蛋白含量与未处理相比差异显著,平均低于未经超声处理的空白组1.28%。综上,超声处理二次溶解的料液不利于蛋白回收率的提高;豆乳是否进行超声处理对酸沉回收率无统计学影响;酸沉后再进行超声处理能够减少大豆乳清中的蛋白损失。
Abstract:The effects of ultrasonic technology on the residual protein content in okara and the protein content in soy whey during soybean protein isolate (SPI) production was investigated. This study investigated the differences in secondary structure between proteins in okara and those in SPI,as well as the impact of ultrasonic treatment during extraction on residual protein content in system effluents (soybean residue and soybean whey). The results showed that,compared with SPI,the secondary structure of okara protein exhibited an 18.83% lower α-helix content,a 13.40% lower β-sheet content,a 9.40% lower random coil content,and a 41.60% higher β-turn content. After 5 min of ultrasonic treatment,the crude protein content in okara increased by 0.4%. For soy whey derived from ultrasonically treated soymilk,there was no correlation between ultrasonication time and the residual protein content in soybean whey. Ultrasonic treatment of the acid-precipitated slurry resulted in a significantly different protein content in the soy whey compared to untreated samples,with an average value 1.28% lower than that of the untreated blank. The findings indicate that ultrasound treatment of the redissolved slurry is unfavorable for improving protein recovery;ultrasonic treatment of soymilk has no statistically significant effect on the recovery rate of acid-precipitated protein;and ultrasound treatment after acid precipitation reduces protein loss in soy whey.
