中文摘要: 为丰富朝鲜蓟的综合利用方法,该研究通过单因素和正交试验,优化超声辅助酶法提取朝鲜蓟可溶性膳食维(soluble dietary fiber,SDF)的提取工艺,并探究该法对朝鲜蓟SDF得率、结构、理化性质和功能特性的影响。结果表明,超声辅助酶法提取朝鲜蓟SDF的最佳工艺条件为料液比1∶30 (g/mL)、混合酶(耐高温α-淀粉酶∶碱性蛋白酶=1∶1,质量比)添加量0.5%、超声功率300 W、超声时间80 min,该条件下朝鲜蓟SDF得率为17.12%。与膳食纤维(dietary fiber,DF)相比,SDF中糖苷键主要以β-构型存在,结晶度降低,孔隙增多,结构较疏松;持水性、持油性、溶解度及抗氧化活性均明显提升。综上,超声辅助酶法提取可以改善朝鲜蓟SDF的结构和功能特性,提升加工性能。
Abstract:To enrich the comprehensive utilization methods of Cynara scolymus L.,the extraction process of soluble dietary fiber (SDF) from Cynara scolymus L.by ultrasonic-assisted enzymatic method was optimized using single-factor and orthogonal experiments.The effects of this method on the yield,structure,physicochemical properties,and functional characteristics of the SDF were investigated.The results showed that the optimal conditions for ultrasonic-assisted enzymatic extraction of SDF were a solid-liquid ratio of 1∶30 (g/mL),a mixed enzyme addition (thermostable α-amylase∶alkaline protease=1∶1,mass ratio) of 0.5%,an ultrasonic power of 300 W,and an ultrasonic time of 80 min.Under these conditions,the yield of SDF from Cynara scolymus L.was 17.12%.Compared with dietary fiber (DF),the glycosidic bonds in SDF mainly existed in β-configuration;the crystallinity decreased;porosity increased,and the structure was looser.Additionally,the waterholding capacity,oil-holding capacity,solubility,and antioxidant activity were significantly improved.In conclusion,ultrasonic-assisted enzymatic extraction can improve the structure and functional characteristics of SDF from Cynara scolymus L.and enhance its processing performance.
