中文摘要: 为探究产四甲基吡嗪(tetramethylpyrazine,TTMP)枯草芽孢杆菌ZP菌株(Bacillus subtilis ZP)的蛋白质和风味代谢的途径、关联及差异,该试验采用顶空固相微萃取结合气相色谱-质谱(headspace solid phase microextraction combined with gas chromatography-mass spectrometry,HS-SPME-GC-MS)法对B.subtilis ZP菌株发酵液的挥发性组分进行检测,并对该菌株进行全基因组、转录组、蛋白质组和风味组的联合解析。结果表明,B.subtilis ZP菌株的基因组由1个Contig序列组成,大小为3.99×106 bp,该菌株经2种不同培养基发酵后筛选到1 010个差异基因和98个差异表达蛋白质,鉴定出40种显著差异代谢产物,控制糖代谢和氨基酸降解等差异通路的相关基因可能对该菌株产TTMP具有调控作用。
Abstract:To explore the pathways,correlations,and differences in protein and flavor metabolism related to tetramethylpyrazine (TTMP) production in Bacillus subtilis (B.subtilis ZP),this study employed headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS) to analyze the volatile components in the fermentation broth.Additionally,an integrated analysis of the whole genome,transcriptome,proteome and flavorome was performed.The results showed that the genome of B.subtilis ZP consisted of a single contig with a size of 3.99×106 bp.Following fermentation in two different media,1 010 differentially expressed genes and 98 differentially expressed proteins were identified,along with 40 significantly differential metabolites.Genes associated with differential pathways,such as those involved in carbohydrate metabolism and amino acid degradation,may play a regulatory role in TTMP production by B.subtilis ZP.
