中文摘要: 以菜籽饼粕和亚麻籽饼粕为原料,利用碱溶酸沉法提取菜籽饼粕蛋白和亚麻籽饼粕蛋白,通过单因素和响应面试验优化菜籽饼粕蛋白和亚麻籽饼粕蛋白提取工艺,并对两种饼粕蛋白的理化特性和功能特性进行对比分析。工艺优化结果表明:菜籽饼粕蛋白提取工艺最佳条件为pH11、浸提时间100 min、浸提温度55 ℃、料液比1∶25 (g/mL),在此条件下菜籽饼粕蛋白提取率为14.27%。亚麻籽饼粕蛋白提取工艺的最佳条件为pH9、一次浸提时间140 min、二次浸提时间60 min、浸提温度60 ℃、料液比1∶20 (g/mL),在此条件下亚麻籽饼粕蛋白提取率为12.96%。品质对比分析结果表明:在理化特性方面,菜籽饼粕蛋白和亚麻籽饼粕蛋白含量分别为72.91%和65.57%,且水分含量均较低,分别为8.39%和6.41%,有利于饼粕蛋白的储存。在功能特性方面,菜籽饼粕蛋白的持水性和持油性较高,分别为30.81%和17.77%,表现出更高的水分保持和保留油脂的能力。菜籽饼粕蛋白和亚麻籽饼粕蛋白的乳化活性和乳化稳定性都较高,分别为78.32%、76.73%、84.19%和81.98%,均具有良好的乳化性能。亚麻籽饼粕蛋白溶解性为69.04%,高于菜籽饼粕蛋白(60.30%)。
Abstract:Rapeseed meal and flaxseed meal were used as raw materials to extract rapeseed meal protein and flaxseed meal protein using the alkali-solution and acid-precipitation method. The extraction processes of both proteins were optimized through single-factor and response surface experiments,and their physicochemical and functional properties were compared and analyzed. The results of process optimization showed that the optimal conditions for rapeseed meal protein extraction were a pH of 11,an extraction time of 100 min,an extraction temperature of 55 ℃,and a solid-to-liquid ratio of 1∶25 (g/mL),under which the extraction rate was 14.27%. The optimal conditions for flaxseed meal protein extraction were a pH of 9,a first extraction time of 140 min,a second extraction time of 60 min,an extraction temperature of 60 ℃,and a solid-to-liquid ratio of 1∶20 (g/mL),under which the extraction rate was 12.96%. Quality comparison showed that,in terms of physicochemical properties,the protein contents of rapeseed meal protein and flaxseed meal protein were 72.91%and 65.57%,respectively,and their moisture contents were low,at 8.39% and 6.41%,respectively,which was favorable for storage. Regarding functional properties,rapeseed meal protein exhibited higher water-holding capacity and oil-holding capacity,at 30.81% and 17.77%,respectively,demonstrating stronger abilities to retain water and oil. Both rapeseed meal protein and flaxseed meal protein showed high emulsifying activity and emulsifying stability,at 78.32%,76.73%,84.19%,and 81.98%,respectively,indicating good emulsifying properties. The solubility of flaxseed meal protein was 69.04%,higher than that of rapeseed meal protein at 60.30%.
