中文摘要: 为制备台蘑调味料,使其游离酸氨基酸含量最大化,以台蘑为原料,探讨不同种类蛋白酶对台蘑酶解效果的影响,筛选酶解效果最佳的蛋白酶,并通过响应面试验优化蛋白酶酶解工艺。结果显示,风味蛋白酶处理后的水解液可溶性固形物含量和呈味氨基酸含量明显高于其他处理组,水解液清透明亮。因此选择风味蛋白酶对台蘑进行酶解处理。采用单因素试验法及响应面分析,以水解溶液中的游离氨基酸浓度为考察指标,对其进行影响因素的研究。结果表明:最佳酶解工艺条件为pH6.4、酶解温度54 ℃、加酶量1 460 U/g、酶解时间3 h,此工艺条件下的风味蛋白酶水解效果最佳,所得到的水解液可以作为台蘑调味品加工基料。
Abstract:To prepare Wutai Mountain mushroom seasoning with maximized content of free amino acid,the effect of different proteases on the enzymatic hydrolysis of Wutai Mountain mushroom was investigated. The protease with the best performance of enzymatic hydrolysis was selected,and its hydrolysis process was optimized through response surface experiments. Results showed that the hydrolysate treated with flavor protease exhibited significantly higher content of soluble solids and flavor amino acids compared to other treatment groups,along with a clear and bright appearance. Therefore,flavor protease was chosen for Wutai Mountain mushroom enzymolysis. By single-factor experiments and response surface analysis,the influencing factors were investigated with the concentration of free amino acids in the hydrolysate as the evaluation index. The results demonstrated that the optimal hydrolysis conditions for flavor protease were as follows: pH of 6.4,temperature of 54 ℃,enzyme dosage of 1 460 U/g,and hydrolysis time of three hours. Under these conditions,flavor protease achieved the best hydrolysis effect. The produced hydrolysate was suitable as a base material for Wutai Mountain mushroom seasoning.
