中文摘要: 该研究以蔓三七叶为原料,经70%乙醇提取得到蔓三七粗提物,粗提物经过石油醚(petroleum ether,PE)脱脂处理得到蔓三七脱脂浸膏和PE组分,采用热溶冷沉的方式对脱脂浸膏进行反复洗涤得到不同含量的绿原酸组分,并探讨其对脂多糖(lipopolysaccharide,LPS)刺激RAW264.7细胞免疫调节活性的影响。结果显示,随着热溶冷沉次数的增加,沉淀中绿原酸含量也在逐渐提高,第6次沉淀物中的绿原酸含量最高,为26.82%,第6次上清液中绿原酸含量为12.06%,回收率达84.81%;抗炎结果显示,蔓三七粗提取物、PE组分以及第1次沉淀物对细胞释放NO水平产生了显著抑制作用且对细胞存活率的影响较小,这3类物质中绿原酸含量分别为7.05%、2.03%和1.35%。综上,采用脱脂后沉降可以为绿原酸提供一个简易、快捷的纯化方法。
Abstract:Using Gynura procumbens as the raw material,this study extracted the crude extract of G. procumbens with 70% ethanol. The crude extract was defatted with petroleum ether (PE) to obtain the defatted extract and PE fraction of G. procumbens. The defatted extract was repeatedly washed by hot dissolution and cold precipitation to obtain chlorogenic acid components with different content. Their immunomodulatory effects on lipopolysaccharide (LPS)-induced RAW264.7 macrophages were investigated. The results showed that with the increase of hot dissolution and cold precipitation times,the chlorogenic acid content in the precipitate gradually increased. The highest chlorogenic acid content was detected in the sixth precipitate,which was 26.82%.The chlorogenic acid content in the sixth supernatant was 12.06%,and the recovery rate was 84.81%. The antiinflammatory results showed that the crude extract,the PE fraction,and the first precipitate of G. procumbens significantly inhibited the secretion of NO and had little effect on cell survival rate. The chlorogenic acid content in these three components was 7.05%,2.03%,and 1.35%,respectively. Overall,sedimentation after defatting provides a simple and fast purification method for chlorogenic acid.
