中文摘要: 该研究使用响应面法优化铜绿假单胞菌的培养基及培养条件,并对优化前后所产鼠李糖脂的性状进行表征。选定碳源浓度、氮源浓度、磷源浓度、初始pH值、摇床转速及接种量作为考察因素,通过Plackett-Burman试验筛选影响鼠李糖脂浓度的显著性因素为氮源浓度、摇床转速及接种量。在此基础上进行Box-Behnken响应面试验,确定3个显著性因素的最佳组合为氮源浓度4.74 g/L、接种量6.22%、摇床转速203.76 r/min,该条件下的鼠李糖脂浓度高达23.70 g/L。此外,对产物进行分离纯化并对比优化前后的鼠李糖脂性能,测试鼠李糖脂的临界胶束浓度及乳化能力。结果表明,以甘油为碳源的优化前培养基所产鼠李糖脂表面活性较高,而以大豆油为碳源的优化后培养基所产鼠李糖脂乳化能力较好。
Abstract:The response surface methodology was employed to optimize the culture medium and conditions of Pseudomonas aeruginosa for enhanced production of rhamnolipids,and the produced rhamnolipids before and after optimization were characterized.The factors studied included concentrations of carbon,nitrogen,and phosphorus sources,initial pH,rotating speed,and inoculum percentage.The significant factors affecting the concentration of rhamnolipids—identified through Plackett-Burman design—were nitrogen source concentration,rotating speed,and inoculum percentage.On the basis of these findings,Box-Behnken response surface experiments determined the optimal conditions as nitrogen source concentration of 4.74 g/L,inoculum percentage of 6.22%,and rotating speed of 203.76 r/min,under which the concentration of rhamnolipids reached 23.70 g/L.Furthermore,the products were purified and characterized,and the critical micelle concentration and emulsifying ability of rhamnolipids were determined.The results indicated that rhamnolipids produced by the unoptimized medium with glycerol as the carbon source exhibited higher surface activity,whereas rhamnolipids produced by the optimized medium with soybean oil as the carbon source showed superior emulsifying ability.
