中文摘要: 为提升分散泛菌(Pantoea dispersa)来源的β-1,4-内切葡聚糖酶(β-1,4-endoglucanase,panEG1)在大肠杆菌BL21(DE3)宿主菌株中的表达效率,该研究采用单因素试验,系统考察诱导温度、诱导持续时间、异丙基-β-D-硫代半乳糖苷(isopropyl β-D-thiogalactopyranoside,IPTG)诱导剂浓度及诱导起始阶段菌液OD600 值这4项关键因素,对PanEG1蛋白表达量的具体影响规律,明确该酶在目标宿主中高效表达的适宜条件,并在最佳诱导条件的基础上进行重组PanEG1的动力学研究。结果表明,重组PanEG1在大肠杆菌BL21(DE3)中的最适表达条件为诱导温度28 ℃、诱导时间20 h、IPTG浓度0.10 mmol/L、诱导前菌液OD600 值为1.6。
Abstract:To enhance the expression efficiency of β-1,4-endoglucanase(panEG1)from Pantoea dispersa in the Escherichia coli BL21(DE3)host strain,a single-factor experimental design was adopted in this study to systematically investigate four key factors:induction temperature,induction duration,isopropyl β-D-thiogalactopyranoside(IPTG)inducer concentration,and pre-induction OD600 value of the culture.The specific effect of each factor on the expression level of PanEG1 enzyme protein was analyzed.The aim was to determine the optimal conditions for the efficient expression of this enzyme in the target host and to conduct enzyme kinetics studies of the recombinant PanEG1 enzyme under the best induction conditions.The results indicated that the optimal expression conditions for recombinant PanEG1 in E.coli BL21(DE3)were:an induction temperature of 28 ℃,an induction duration of 20 h,an IPTG concentration of 0.10 mmol/L,and a pre-induction OD600 value of the bacterial solution of 1.6.
