中文摘要: 该研究旨在制作一款黄精乌鸡汤,并结合感官评价、网络药理学和动物实验探究其免疫调节作用机制。首先利用数据库筛选黄精乌鸡汤与免疫力交集靶点并进行富集分析,其次制备黄精乌鸡汤并进行感官评价,最后构建环磷酰胺(cyclophosphamide,CTX)诱导的免疫抑制小鼠模型,并灌胃不同剂量的黄精乌鸡汤冻干粉水溶液,评估其对小鼠免疫功能的影响。结果显示,共筛选出206个黄精乌鸡汤提高免疫力的潜在作用靶点,富集于趋化因子、T细胞受体、磷脂酰肌醇3激酶等信号通路。感官评价结果表明,制作所得的黄精乌鸡汤汤体澄清透亮,呈浅琥珀色,香气浓郁,汤鲜味浓且回甘。动物实验结果显示,与正常(Control)组相比,模型(CTX)组小鼠相对体质量极显著下降,同时脾脏指数、胸腺指数及血清免疫球蛋白A(immunoglobulin A,IgA)、免疫球蛋白G(immunoglobulin G,IgG)、免疫球蛋白M(immunoglobulin M,IgM)水平和血常规指标[白细胞(white blood cell,WBC)、红细胞(red blood cell,RBC)、血红蛋白(hemoglobin,HGB)和淋巴细胞(lymphocyte,LYMPH)]水平均显著降低(P<0.05,P<0.01)。而黄精乌鸡汤各组均能不同程度缓解CTX诱导的相对体质量下降,并显著提升上述免疫器官指数、免疫球蛋白水平及血常规指标。
Abstract:This study aimed to prepare a Polygonati Rhizoma and black bone chicken soup,and investigate its immunomodulatory effect and mechanism through integrated sensory evaluation,network pharmacology,and animal experiment.First,potential targets associated with the immunomodulatory effects of the soup were screened from the database,followed by enrichment analysis.Then,the soup was prepared and subjected to sensory evaluation.Finally,an immunosuppressed mouse model induced by cyclophosphamide(CTX)received different doses of freeze-dried powder solution of the soup by gavage to assess its effect on immune function.According to the results,a total of 206 potential targets were identified to be related to the immune-enhancing effect of the soup,which were enriched in signaling pathways such as the chemokine,T-cell receptor,and phosphatidylinositol 3-kinase(PI3K-Akt)pathways.Sensory evaluation indicated that the prepared soup was transparent and light amber,characterized by robust aromatic properties,dominant umami flavor,and a distinctive sweet aftertaste.In the animal experiments,compared to the control group,the CTX group exhibited a significantly decreased relative body weight,along with decreased spleen and thymus index,serum immunoglobulin(IgA,IgG,and IgM)levels,and hematological parameters [white blood cells(WBC),red blood cells(RBC),hemoglobin(HGB),and lymphocytes(LYMPH)](P<0.05,P<0.01).In contrast,intervention with different doses of the soup alleviated the decrease in relative body weight to varying degrees and significantly increased the aforementioned immune organ index,immunoglobulin levels,and hematological markers.
