不同地区薤白中降胆固醇乳酸菌的分离筛选及益生功能评价

中文摘要: 该研究从不同地区薤白中分离乳酸菌,对菌株进行筛选鉴定,并对筛选出的菌株益生功能进行评价。结果表明,共分离筛选得到173株革兰氏阳性菌株,体外评价其降胆固醇、耐酸和耐胆盐能力,其中菌株HBXB-19、GZXB-22、GZXB-28和JXXB-18性能良好,其对胆固醇的降解率均大于50%,在pH2的溶液中处理4 h的存活率均大于90%,在含有0.3%牛胆盐的溶液中处理4 h的存活率均大于70%,经16S rRNA序列分析,4株菌株均为植物乳植杆菌(Lactiplantibacillus plantarum)。对4株菌株的益生功能评价结果表明:菌株GZXB-28的生长性能最好,其培养24 h后在600 nm下吸光度达到2.485;菌株GZXB-28耐胃肠液能力较好,其在人工胃液中处理2 h的存活率为80.85%,在人工肠液中处理2 h的存活率为55.60%;菌株GZXB-22和GZXB-28具有较强的疏水性,其疏水性分别为46.28%和54.64%,菌株GZXB-28和JXXB-18具有较强自聚性,其自聚性分别为58.32%和62.43%;菌株GZXB-22、GZXB-28和JXXB-18均对4种致病菌有抑制作用,其中菌株GZXB-22对沙门氏菌C79-13和蜡样芽孢杆菌ATCC 14579有最强的抑制作用,菌株GZXB-28对金黄色葡萄球菌ATCC 25923和大肠杆菌ATCC 25922有最强的抑制作用;4株菌株均对青霉素、氨苄西林、红霉素、四环素、万古霉素、氯霉素、阿莫西林和头孢唑林表现为不耐受,安全性良好。综上,植物乳植杆菌GZXB-28的各项性能优良,可作为开发新型降胆固醇产品的潜力菌株。

Abstract:Lactic acid bacteria were isolated from Allium macrostemon in different regions,and the strains were screened and identified;the probiotic functions of the screened strains were evaluated.The results showed that a total of 173 Gram-positive strains were isolated and screened,and their cholesterol-lowering,acid-resistant,and bile salt-resistant abilities were evaluated in vitro. Among them,strains HBXB-19,GZXB-22,GZXB-28,and JXXB-18 had good performance,and their cholesterol degradation rates were all greater than 50%.The survival rates after 4 h treatment in pH2 solution were all greater than 90%,and the survival rates after 4 h treatment in solution containing 0.3% bovine bile salt were all greater than 70%. The 16S rRNA sequence analysis showed that the four strains were all Lactiplantibacillus plantarum. The results of probiotic function evaluation of the four strains showed that the growth performance of strain GZXB-28 was the best,and the absorbance at 600 nm reached 2.485 after 24 h of culture. Strain GZXB-28 had good tolerance to gastrointestinal fluid. The survival rates of strain GZXB-28 was 80.85% after 2 h treatment in artificial gastric fluid and 55.60% after 2 h treatment in artificial intestinal fluid. Strains GZXB-22 and GZXB-28 had strong hydrophobicity,with hydrophobicity of 46.28% and 54.64%,respectively. Strains GZXB-28 and JXXB-18 had strong self-aggregation,with self-aggregation of 58.32% and 62.43%,respectively. Strains GZXB-22,GZXB-28,and JXXB-18 all had inhibitory effects on four pathogenic bacteria. Among them,strain GZXB-22 had the strongest inhibitory effect on Salmonella C79-13 and Bacillus cereus ATCC 14579,and strain GZXB-28 had the strongest inhibitory effect on Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922.The four strains were all intolerant to penicillin,ampicillin,erythromycin,tetracycline,vancomycin,chloramphenicol,amoxicillin,and cefazolin,and the safety was good.In summary,Lactobacillus plantarum GZXB-28 had excellent properties and could be used as a potential strain for the development of new cholesterol-lowering products.
