中文摘要: 该研究以小龙虾壳为原料,探究氢氧化钠/尿素冻融结合高压蒸煮对小龙虾虾壳甲壳素提取的影响,结果表明冻融3次结合高压蒸煮(100 kPa、121 ℃、1 h)可制得纯度为99.02%的甲壳素。以其为底物,利用甲壳素脱乙酰酶催化反应制备壳聚糖,在反应时间12 h、甲壳素脱乙酰酶添加量1.0 U/mL、底物浓度1 mg/mL的条件下所制备的壳聚糖脱乙酰度达到92.62%。此外,通过扫描电子显微镜(scanning electron microscopy,SEM)、傅里叶变换红外光谱(Fourier transform-infrared spectroscopy,FT-IR)和X-射线衍射(X-ray diffraction,XRD)对甲壳素、壳聚糖进行表征,结果表明甲壳素结构松散,有利于酶促反应,且制备工艺会对壳聚糖结构产生影响。该方法可以避免大量碱液的使用,且酶促反应过程温和,所得壳聚糖的脱乙酰度较高。
Abstract:Crayfish shells were utilized as the raw material for the extraction of chitin through sodium hydroxide/urea freeze-thawing combined with high-pressure steaming. The results showed that three cycles of freezethawing combined with high-pressure steaming(100 kPa,121 ℃,1 h)yielded chitin with the purity of 99.02%. The obtained chitin was used as the substrate for the preparation of chitosan via a reaction catalyzed by chitin deacetylase. After reaction with 1.0 U/mL chitin deacetylase and 1 mg/mL substrate for 12 h,the deacetylation degree of the obtained chitosan reached 92.62%. Furthermore,chitin and chitosan were characterized by scanning electron microscopy(SEM),Fourier transform-infrared spectroscopy(FTIR),and X-ray diffraction(XRD).The results indicated that chitin had a loose structure,which facilitated the enzymatic reaction,and the preparation process affected the structure of chitosan. This approach avoided the use of large amounts of alkaline solution,had mild enzymatic reaction conditions,and produced chitosan with a high deacetylation degree.
