中文摘要: 为优化黄变稻谷总黄酮的提取工艺并评价其抗氧化活性,以黄变稻谷为原料,采用超声辅助提取法,以总黄酮提取率为评价指标,考察不同原料预处理方式(吸水涨润研磨、液氮研磨机研磨、高速粉碎机研磨)及不同提取溶剂(水、甲醇、乙醇、乙酸乙酯)对提取效果的影响。结果表明:液氮研磨机研磨为原料最优预处理方式,乙醇为最佳提取溶剂。进一步通过单因素试验及Box-Behnken响应面法,得到最佳提取条件为提取温度50 ℃、料液比1∶58(g/mL)、乙醇体积分数83%、摇床时间46 min、超声时间69 min,此条件下总黄酮提取率为9.66%。黄变稻谷展现出良好的抗氧化活性,黄酮提取物的抗氧化能力显著高于未黄变稻谷。
Abstract:To optimize the extraction process of total flavonoids from yellowed rice and evaluate their antioxidant activity,with yellowed rice as the raw material,an ultrasound-assisted extraction method was employed.The extraction rate of total flavonoids was used as the evaluation index to investigate the effects of different raw material pretreatment methods(swelling and grinding,grinding with a liquid nitrogen grinder,and grinding with a high-speed crusher)and different solvents(water,methanol,ethanol,and ethyl acetate)on the extraction effects. The results indicated that grinding with a liquid nitrogen grinder was the optimal pretreatment method,and ethanol was the best extraction solvent. Furthermore,through single-factor experiments and Box-Behnken response surface methodology,the optimal extraction conditions were determined as follows:extraction temperature of 50 ℃,solid-liquid ratio of 1∶58(g/mL),ethanol volume fraction of 83%,shaking time of 46 min,and ultrasonic time of 69 min. Under these conditions,the extraction rate of total flavonoids reached 9.66%.The yellowed rice demonstrated good antioxidant activity,and the antioxidant properties of its flavonoid extract were significantly higher than those of non-yellowed rice.
