中文摘要: 为促进米渣蛋白的高值化利用及植物蛋白发泡剂的开发,以米渣蛋白为材料,比较pH循环碱法水解、pH循环酸法水解及酶法水解对米渣蛋白起泡特性的影响,并解析其改性机理。通过单因素试验分别优化得出3种方法的最佳工艺条件。结果表明,pH循环碱法水解(80 ℃水解3 h,pH10)对米渣蛋白起泡性及泡沫稳定性的提升效果最为显著,分别达300%和73%,优于酶法水解及pH循环酸法水解处理。结构表征显示,改性后蛋白粒径减小、分子量分布改变、二级结构重排(α-螺旋与无规卷曲增加)、表面疏水性降低,且pH循环碱法水解处理中钙离子的交联作用有助于形成稳定泡沫结构。
Abstract:The value-added utilization of rice dreg protein and the development of plant-based foaming agents were explored.Rice dreg protein was used as the raw material for comparison of the effects of pH-shift alkaline hydrolysis,pH-shift acid hydrolysis,and enzymatic hydrolysis on the protein foaming properties,and the corresponding modification mechanisms were elucidated.Single-factor experiments were conducted to optimize the process conditions for each of the three methods.The results showed that pH-shift alkaline hydrolysis(at 80 ℃ for 3 h,pH10)significantly improved the foaming capacity and foam stability of rice dreg protein(reaching 300% and 73%,respectively),outperforming both enzymatic hydrolysis and pH-shift acid hydrolysis.Structural characterization revealed that the modified protein exhibited a reduced particle size,altered molecular weight distribution,a rearranged secondary structure(increased α-helix and random coil content),and decreased surface hydrophobicity.Additionally,the cross-linking effect of calcium ions during pH-shift alkaline hydrolysis contributed to the formation of a stable foam structure.
