中文摘要: 类布氏乳杆菌(Lentilactobacillus parabuchneri) C41是一株从番茄发酵液分离的产核黄素菌株,该文通过全基因组测序分析探究C41菌株的产核黄素机制和潜在危害。采用Pacbio加Illumina测序的方法获取C41菌株全基因组序列,通过编码基因预测和基因注释等生物信息学方法分析菌株的有关功能机制和潜在危害性。结果显示,C41菌株基因组含1个环状DNA和3个质粒,环状DNA大小为2 650 018 bp,GC含量为43.53%,质粒A、质粒B和质粒C的大小分别为58 659、50 453 bp和49 016 bp,预测得到2 802个编码基因,64个tRNA基因,15个rRNA基因。通过KEGG注释发现C41菌株染色体包含核黄素从头合成所需酶的编码基因。碳水化合物活性酶与次级代谢产物合成基因分析共注释到66个碳水化合物活性酶编码基因、1个III型聚酮合酶基因簇和2个核糖体合成和翻译后修饰肽基因簇。致病菌毒力因子数据库与综合抗生素耐药性数据库比对分析显示C41基因组没有基因被注释为毒力基因,1个基因被注释为抗菌药物耐药相关基因。结果表明C41菌株具有从头合成核黄素的能力和较好的安全性,具备进一步开发应用的潜力。
Abstract:Lentilactobacillus parabuchneri C41 is a riboflavin-producing strain isolated from tomato fermentation broth.This study employed whole genome sequencing and analysis to explore the mechanism of riboflavin production and the potential hazards of the C41 strain.The whole genome sequence of the C41 strain was obtained by Pacbio and Illumina sequencing.The functional mechanism and potential risks of the strain were analyzed by coding gene prediction and annotation.The results showed that the genome of strain C41 contained circular DNA and three plasmids.The size of the circular DNA was 2 650 018 bp,and the GC content was 43.53%.The sizes of plasmid A,plasmid B,and plasmid C were 58 659 bp,50 453 bp,and 49 016 bp,respectively.2 802 coding genes,64 tRNA genes,and 15 rRNA genes were predicted.Through KEGG annotation,it was discovered that the chromosome of strain C41 contained the enzyme-encoding genes required for the de novo synthesis of riboflavin.Furthermore,the analysis of carbon-hydrocarbon active enzymes and secondary metabolite synthesis genes jointly contained 66 carbon-hydrocarbon active enzyme coding genes,one type III polyketide synthase gene cluster,and two ribosomally synthesized and post-translationally modified peptide gene clusters.The comparison analysis between the pathogenicity factor database of pathogenic bacteria and the comprehensive antibiotic resistance database revealed that no genes in the C41 genome were annotated as virulence genes,while one gene was annotated as a gene related to antibiotic resistance.The results demonstrated that the C41 strain possessed the capacity for the de novo synthesis of riboflavin and exhibited favorable safety profiles,rendering it promising for further research.
