中文摘要: 为比较冠突散囊菌(Eurotum cristatum)固态发酵前后荷叶活性成分及功能变化,该文对发酵前后荷叶活性成分及体外抗氧化、降血脂功能进行比较分析,明确各成分含量与活性之间的相关性。结果表明:与发酵前相比,发酵后荷叶提取物总多糖含量在发酵2 d显著提升[(62.62±0.87)mg/g干荷叶],总黄酮含量在发酵18 d显著增多[(47.10±1.69)mg/g干荷叶]。DPPH·和ABTS+自由基清除能力、总还原能力结果表明,相比于发酵前,发酵后16 d和18 d荷叶抗氧化活性有所增强。胆酸盐结合能力和胰脂肪酶抑制能力结果表明,发酵16、22 d的荷叶提取物的胆酸盐结合能力和胰脂肪酶抑制能力较好,表明发酵可以在一定程度上增强降血脂活性。相关性分析结果表明,总黄酮、总多糖、总多酚、总生物碱、总蛋白含量与抗氧化活性、甘氨胆酸钠结合能力和牛磺胆酸钠结合能力呈极显著正相关(P<0.01)。综上,冠突散囊菌发酵可以增强荷叶体外抗氧化、降血脂活性,其中发酵16、18、22 d的荷叶提取物的相关功能活性较强。
Abstract:To compare the changes in active components and functions of lotus leaves before and after solidstate fermentation by Eurotum cristatum,this study analyzed variations in active components as well as in vitro antioxidant and hypolipidemic activities before and after fermentation,thus clarifying the correlation between component content and bioactivities.The results showed that compared with unfermented samples,the total polysaccharide content of fermented lotus leaf extracts increased significantly after 2 days of fermentation[(62.62±0.87) mg/g dry weight],while the total flavonoid content was significantly increased after 18 d of fermentation [(47.10±1.69) mg/g dry weight].Results from DPPH· and ABTS? free radical scavenging and total reducing power assays indicated that the antioxidant activity of lotus leaves was enhanced after 16 and 18 days of fermentation compared to the unfermented samples.The bile salt binding capacity and pancreatic lipase inhibition assay indicated improved results in lotus leaf extracts fermented for 16 d and 22 d,suggesting that fermentation could enhance hypolipidemic activity to some extent.Correlation analysis revealed that the total content of flavonoids,polysaccharides,polyphenols,alkaloids,and protein was significantly positively correlated with antioxidant activity,sodium glycocholate binding capacity,and sodium taurocholate binding capacity (P<0.01).In conclusion,solid-state fermentation by Eurotum cristatumcan enhanced the in vitro antioxidant and hypolipidemic activities of lotus leaves,with stronger functional activities in extracts fermented for 16,18 d,and 22 d.
