中文摘要: 建立蛋白质水解配方食品、水解蛋白粉中分子量及其分布范围的凝胶色谱检测方法,以添加0.05%的三氟乙酸乙腈水混合溶液为提取剂,采用TSKgel G2000 SWXL凝胶色谱柱,在流动相为乙腈∶水∶三氟乙酸=40∶60∶0.05(体积比)、波长 221 nm的条件下进行分离检测,通过建立不同分子量的蛋白标准品校正曲线,测定样品中分子量和分子量分布。结果 显示,蛋白标准品分子量在189~12 384 Da时线性关系良好,标准曲线方程为y=-0.357 2x+7.115 3,相关系数R2=0.99,样品检测试验重复性良好,各肽段的分子量分布相对标准偏差均小于10%。结果表明,该方法分析速度快,简单易行、精密度较高,重复性良好,能快速测定蛋白质水解配方食品、水解蛋白粉中相对分子量及其分布。
Abstract:A gel chromatography (GPC) method was established for the determination of the molecular weights and their distributions in protein hydrolyzed formula foods and hydrolyzed protein powder. The samples were extracted with a solvent composed of acetonitrile/water containing 0.05% trifluoroacetic acid. The separation was then carried out on a TSKgel G2000 SWXL gel filtration column under the following conditions: a mobile phase of acetonitrile∶water∶trifluoroacetic acid = 40∶60∶0.05 and a detection wavelength of 221 nm. The molecular weight and its distribution were determined by establishing a calibration curve using protein standards of different molecular weights. The results showed that the linear relationship was good when the molecular weight of the protein standard sample was between 189 Da and 12 384 Da. The standard curve equation was y=-0.357 2x+7.115 3, and correlation coefficient R2=0.99, with good repeatability. The relative standard deviation of the molecular weight distribution of each peptide was less than 10%. The results indicated that the method is characterized by its fast analysis speed, simplicity and ease of operation, high precision, good repeatability, and the ability to quickly determine the relative molecular weight and distribution of protein hydrolyzed formula foods and hydrolyzed protein powder.
