中文摘要: 为分析黄芪中总砷及4种砷形态的含量范围,建立液相色谱-原子荧光(liquid chromatography-atomic fluorescence spectrometry, LC-AFS)法测定黄芪中4种砷形态含量的方法。采用微波消解法消解黄芪样品,原子荧光法测定黄芪中总砷的含量;利用Box-Behnken试验,确定黄芪中砷形态化合物提取的最优条件。结果表明,30份黄芪样品中总砷的检出率为50%,含量范围在ND~0.35 mg/kg。经优化后,黄芪中砷形态化合物提取的最佳条件为硝酸浓度0.20 mol/L、提取温度62.2 ℃、提取时间为35.6 min,此时实际提取率为89.3%,与预测值差异较小。利用LC-AFS分离黄芪样品中4种砷形态并进行测定,在0~100 μg/L范围内4种砷形态化合物在20.0 min内得到有效分离,各形态相关系数均达到0.999以上。

Abstract:To analyze the content range of total arsenic and four arsenic forms in Astragali Radix, a content determination method was established based on liquid chromatography-atomic fluorescence spectrometry (LCAFS). The microwave digestion method was used to digest Astragali Radix samples, and the atomic fluorescence method was employed to determine the total arsenic content in Astragali Radix. The conditions for extracting arsenic compounds from Astragali Radix were optimized by the Box-Behnken design. The detection rate of total arsenic in 30 Astragali Radix samples was 50%, with a content range of ND-0.35 mg/kg. The optimal conditions for extracting arsenic compounds from Astragali Radix were a nitric acid concentration of 0.20 mol/L, an extraction temperature of 62.2 ℃, and extraction time of 35.6 min, under which the extraction rate reached 89.3%, which showed a minor difference from the predicted value. LC-AFS was used to separate four arsenic forms from Astragali Radix samples for determination. The four arsenic forms in the range of 0-100 μg/L were effectively separated within 20.0 min. The correlation coefficients of different forms were above 0.999.
