中文摘要: 为探讨广式老火汤中常见食药材对黄嘌呤氧化酶(xanthine oxidase,XO)的抑制作用,采用75%乙醇和水分别提取8种常见食药材,测定其粗提物提取率及对XO活性的抑制能力,并研究不同提取物对XO的抑制机理。结果表明,7种食药材的水提物提取率高于醇提物,其中土茯苓和木棉花的醇提物与水提物提取率差异较小,提取率较为稳定;16种粗提物对XO的半数抑制浓度(half maximal inhibitory concentration,IC50)为124.27~2 232.00 μg/mL。抑制机理研究表明,土茯苓醇提物、土茯苓水提物和木棉花水提物为XO的以竞争性抑制为主的可逆性抑制剂,而木棉花醇提物为竞争性可逆抑制剂。
Abstract:To explore the inhibitory effect of common edible medicinal materials on xanthine oxidase (XO) in Cantonese Laohuo soup, this paper extracted eight common edible medicinal materials with 75% ethanol and water, determined the extraction rate of their crude extracts and the ability to inhibit the activity of XO, and investigated the inhibitory mechanism of different edible medicinal material extracts on XO. The results showed that the extraction rate of the aqueous extracts from seven edible medicinal materials was greater than that of the alcoholic extracts. The alcoholic and aqueous extracts of Smilax glabra Roxb. and the alcoholic and aqueous extracts of Bombax ceiba Linn. were more stable, and the difference in the yield of the two crude extracts was smaller than that of the other edible medicinal materials. The half maximal inhibitory concentration(IC50) of the crude extracts of the 16 edible medicinal materials was in the range of 124.27-2 232.00 μg/mL;the alcoholic and aqueous extracts of Smilax glabra Roxb. and the aqueous extract of Bombax ceiba Linn. were reversible inhibitors primarily based on competitive inhibition for XO, and the alcoholic extract of Bombax ceiba Linn. was a competitive reversible inhibitor.
