中文摘要: 为建立一种绿色高效的金花茶总黄酮提取方法,该文采用超声辅助低共熔溶剂(deep eutectic solvents,DESs)技术金花茶总黄酮,以总黄酮得率为响应值,在单因素试验基础上,结合Plackett-Burman试验及Box-Behnken响应面法优化超声辅助低共熔溶剂提取金花茶总黄酮工艺。结果表明:选取乙二醇/氯化胆碱为提取DESs,最佳提取工艺为乙二醇与氯化胆碱物质的量比4∶1、超声功率180 W、提取温度70 ℃、含水量40%、液料比41∶1(mL/g)、超声时间51 min,在此条件下,金花茶总黄酮得率为(10.65±0.26)%,清除DPPH自由基、ABTS+自由基的IC50值为32.91、221.5 μg/mL,优于甲醇提取物。因此,超声辅助低共熔溶剂可绿色、高效地提取金花茶总黄酮。
Abstract:To establish a green and highly efficient extraction process of total flavonoids from Camellia petelotii,ultrasound-assisted deep eutectic solvents (DESs) technology was employed to extract total flavonoids from Camellia petelotii. With the yield of total flavonoids as the response value,the ultrasonic-assisted DESs extraction process of total flavonoids from Camellia petelotii was optimized by combining the Plackett-Burman experiment and the Box-Behnken response surface method. The results showed that when using ethylene glycol/choline chloride as the extraction agent for DESs,the optimal extraction process was as follows:the molar ratio of ethylene glycol to choline chloride was 4∶1,the ultrasonic power was 180 W,the extraction temperature was 70 ℃,the moisture content was 40%,the liquid-to-solid ratio was 41∶1 (mL/g),and the ultrasonic time was 51 min. Under these conditions,the total flavonoid yield from Camellia petelotii was (10.65±0.26)%. The IC50 values for scavenging DPPH free radicals and ABTS+ free radicals were 32.91 and 221.5 μg/mL,respectively,which were superior to those of the methanol extract. Therefore,ultrasonic-assisted DESs can efficiently extract the total flavonoids from Camellia petelotii in a green manner.
